    Identification of a novel SAM-deletion mutation of the receptor tyrosine kinase EPHA3 in liver cancer

    tEPHA3 is a member of the ephrin receptor family, which composes the largest subgroup of the known receptor tyrosine kinases. EPH receptors and their ephrin ligands are essential for a variety of biological processes such as cell adhesions and repulsion, cell migration, axon guidance and angiogenesis et al, and they are implicated in cancer metastasis and invasion. In this study, a novel mutation of Epha3 gene was unexpectedly discovered in liver cancer, where its cytoplasmic SAM domain was incomplete. It is supposedly a novel way for Epha3 regulation in liver cancer.

    Research on function of transcription factor FOXA1 in breast cancer subtyping

    Breast cancer is a highly heterogeneous disease. Its accurate molecular subtyping helps us to improve the precision of breast cancer prognosis and the corresponding therapeutic decision making. By comparing the transcriptional and translational expression of foxa1 among breast cancer cell lines, the previous hypothesis that FOXA1 is a critical factor discriminating breast cancer subtypes was validated. In addition, by silencing foxa1 and observing the subsequent proliferation and apoptosis, the roles of FOXA1 played in breast cancer occurrence, development and prognosis were explored. Analyzing the expression of foxa1 in 10 cell lines covering four subtypes by Western Blot and real-time quantitative PCR, the expression of foxa1 in Luminal A and Luminal B breast cancers was significantly higher than that in Her2 positive and Triple negative subtypes. By analyzing the expression of other genes that promote cell growth, the result showed that FOXA1 was a key regulatory factor between GATA3 and ER, which inhibited cell proliferation.

    Influences of magnetic immunochromatographic assay for detection of PreS1 region of hepatitis B virus surface antigen

    To establish a rapid assay for HBV, a magnetic immunochromatographic assay (MICA) for detection of PreS1Ag in serum samples of people was developed in our study. The influences of hydrodynamic size of magnetic nanobeads, coupled antibody, pH, basal buffer, reaction temperature on the MICA were analyzed emphatically. Results showed that dispersion stability of magnetic nanobeads monitored by hydrodynamic size had a strong effect on MICA. Moreover, optimal antibody addition, proper pH value of 6.5 to 9 and using PBS with more stable pH as the basal buffer could be beneficial to improve the sensitivity and stability of MICA. The results of this research are expected to provide the theoretical guidance for the future development of MICA.

    Overexpression of artificial sRNA anti-micA enhancing Escherichia coli viability and pullulanase production

    Escherichia coli is frequently influenced by various stresses in the process of exogenous protein expression and established various stress signaling systems that sensing and responding to specific stimuli. Recently it has been identified that many small non-coding RNAs (sRNA) have an important role in regulating mRNA translation by influencing the stability and translation efficiency of mRNA. In this research, in order to block sigma E-mediated cell lysis signal pathway, we studied the effect of overexpressing anti-micAs on the tolerance of E. coli to ethanol, pullulanase production and the trans-coded RNA interference efficiency, by using artificial trans-coded sRNAs of micA anti-micAs (bprO and omU) to silence micA. Moreover, the influencing factors of function of artificial trans-coded sRNAs were also discussed. Research found that in bprO and omU overexpression strains, the colony forming units (CFU) were raised by half and one order of magnitude, respectively; the ompA mRNA levels were improved by 7.2-fold and 6.9-fold, respectively and the content of OmpA protein was also significantly increased; pullulanase production was enhanced by 1.4-fold and 1.8-fold, respectively. Moreover, the effect of omU overexpression on Escherichia coli was slightly larger than that of bprO.

    Study on secretory expression of recombination pullulanase from Bacillus acidopullulyticus in Escherichia coli

    The pullulanase of Bacillus acidopullulyticus has low secretory expression level as a macromolecule. For pullulanase secretory in recombination Escherichia coli BL21(DE3), BL21(DE3)-pET-28a(+)-pelB-pul13A with PelB signal peptide was constructed, then fermentation optimization and medium additives were employed for better secretory. Through the construction of recombinant strain BL21(DE3) with pelB, pullulanase was secreted at a low level. By fermentation optimization including medium , induced temperature , the concentration of IPTG, induced time and medium additives, the extracellular pullulanase activity was markedly increased. pullulanase was secreted into periplasmic space. Under the condition of 0.1 mmol/L IPTG, 20℃ induced after D600 nm was about 5.0-6.0, the periplasmic pullulanase activity reached to 102.5 U/mL with addition of 0.03%SDS after induction for 20 h.

    The study of atrazine degraded by endophytic fungus from sugarcane

    An endophytic fungus strain of degrading atrazine was isolated from the roots of sugarcane in Guangxi farmland which has been sprayed by atrazine for a long time. It was preliminarily identified as Fusarium fujikuroi according to its morphological characteristics and the analysis of its 18S rRNA and ITS gene sequences. The degradation characteristics were also investigated. The results showed that the strain could use atrazine as sole nitrogen source and had the best degradation property under those conditions: pH 7-8, temperature 28℃, atrazine primary concentration 25 mg/L. The degradation rate could reach 49.6% after 10 days. The degradation products were determined by ultra performance liquid chromatography-mass spectrometry (UPLC-MS). The results showed that the degradation product was cyanuric acid.

    In this study, the large yellow croakers Donghai No.1 was divided into cold-tolerant and cold-sensitive two groups by reducing water temperature from 15℃ to 6℃ gradually, and then the molecular marker which linked to the resistance to low temperature was screened by microsatellite markers (SSR) analysis. The results showed that 39 alleles were amplified using 10 SSR primers, and only the alleles frequence of locus KPC43 had significant difference between the cold-tolerant group and the cold-sensitive group. In the three alleles of KPC43, the occurrence frequency of KPC43170bp was 80.0% in the cold-tolerant group, while its occurrence frequency was only 23.33% in the cold-sensitive group. Correlative analysis showed that there was a significant positive correlation between KPC43170bp and cold-tolerant trait (P<0.01), and the correlation coefficient was 0.498. In the cold-sensitive group, the frequency of the KPC43150bp (76.67%) was significantly higher than that in the cold-tolerant group (30.0 %). And there was a significant negative association with the KPC43150bp and cold-tolerant related character (P<0.01), and the correlation coefficient was-0.417. Sequence alignment showed that the microsatellite flanking regions of KPC43170bp and KPC43150bp were the same, while the repeat sequence was (TG) 12 in KPC43170bp and (TG) 2 in KPC43150bp. It can be speculated that SSR marker KPC43 may be linked to some cold tolerant gene of large yellow croaker, and the repeat numbers of (TG) may affect these gene′s expressing activity, which is associated to the ability of cold tolerance.

    In order to investigate the role of Neuropeptide Y (NPY), Agouti Aelated Peptide (AgRP), Pro-opiomelanocortin (POMC), Cocaine and Amphetamine Regulated Transcript (CART) peptide expression in hypothalamic neuropeptide in body mass regulation in Tupaia belangeri under different photoperiods, body mass, body fat mass, food intake, serum leptin levels and hypothalamic neuropeptide expression were measured. It showed that short photoperiod increased body mass and body fat mass, and increased food intake. But serum leptin levels showed no significant differences between two groups, and leptin levels showed a positive correlation with body fat mass. Hypothalamic neuropeptide NPY had significant difference, but AgRP, POMC and CART expression had not significant differences between two groups. All of the results suggested that short photoperiod can increase the body mass, body fat mass and food intake. Leptin may play an regulation on body mass regulation by acting on hypothalamic neuropeptide of NPY expression in T. belangeri.

    Bioaccumulation of mercury in Anodonta from the important estuary wetlands in Chaohu Lake

    The content and the space distribution of mercury in Anodonta, a representative freshwater shellfish from the main estuary wetlands in Chaohu Lake, were investigated in this study. The results showed that the sediments from the estuary wetlands had been affected by mercury pollution. Especially the pollution in estuary wetlands of Shiwuli River and Nanfei River was most serious. The accumulation of mercury and methyl mercury in Anodonta was obvious and the contents of them were significantly correlated with the contents in sediments. The freshwater shellfish Anodonta was used as an indicator for monitoring the mercury pollution in the estuary wetland. The accumulation of mercury in various tissues had obvious selectivity (mantle>guts>gill and muscle). The shellfish evaluation results showed that the contents of mercury and methyl mercury in Anodonta were lower than the limited standards and had a lower risk for eating. Anodonta in estuary wetlands should be divided into the conservation areas for ecological protection and resource utilization.

    The research analyzed the biomass distribution of uncultivated lysimachia clethroides and the absorbing ability, migration and correlation between the content of selenium and mineral elements; the samples of uncultivated lysimachia clethroides were gathered to measure the biomass. The contents of mineral elements and selenium were determined respectively by the atomic luminescence chromatography and the atomic fluorescent method,and the correlation analysis was done meantime. The result showed the biomass distribution rates in the leaves of the lysimachia clethroides were the highest, and the content of selenium was negatively correlated to potassium in the roots and fruits. Besides, the leaves possessed the highest content of selenium.

    With the increasing concerns of global climate change caused by the consumption of fossil fuels, much efforts have been spent on the exploration of renewable energy sources in the last couple of decades. Biogas produced from the treatment of organic waste materials is one of the renewables that have attracted a lot of attentions. This paper provides a summary of the studies on thermophilic anaerobic digestion or co-digestion of various organic waste materials for biogas production. Discussions in this paper include main advantages of thermophilic anaerobic digestion technology over mesophilic one, factors that critically impact the efficiency of the anaerobic digestion such as temperature, pH, total solids, carbon/nitrogen ratio, and hydraulic retention time, the effect of the pretreatment of lignocellulosic materials on the biogas production from anaerobic co-digestion of animal manure and agricultural residues, and mathematical models describing the anaerobic digestion processes.

    Scombridae fishes, as economically important marine species, are abundant sources of food proteins. Their comprehensive utilization rate, however, is still at a low level. Bioactive peptides with various biological activities such as antioxidation, anti-hypertension, immunization and antibacterial prepared by enzymatic hydrolysis is an efficient way for higher added-value of Scombridae fishes. This paper reviewed the conditions of enzymatic hydrolysis of Scombridae proteins, the approaches for purification and identification as well as the primary physiological functions of Scombridae peptides. Furthermore, the problems and corresponding countermeasures were discussed and potential applications of bioactive peptides from hydrolysates of Scombridae fishes were summarized.

    In the face of energy crisis, using microalgae to produce biofuel and high value-added products has attracted great attention. Microalgae constitute a diverse group of microorganisms with advantages like fast and efficient growth. In addition, they do not compete for arable land and offer very high lipid yield potential. Microalgae are currently considered as the most promising alternative sources for the next generation of food, feed, cosmetics and renewable energy in the form of biofuel. Major challenges for the development of this resource are to select lipid-rich strains using high-throughput staining for neutral lipid content in microalgae species. Compared with traditional quality method, the fluorescent staining method is simpler, cheaper and easier to operate. In this article, the properties of fluorescent Nile red and BODIPY and the use of fluorescent for lipid measurement in microalgae were summarized.

    CD44, a kind of transmembrane receptor glycoprotein, belongs to adhesion molecular family, and it can be divided into standard CD44 (CD44s) and variant CD44 (CD44v) according to the expression of CD44 exon. Numerous researches confirmed that the abnormal expression of CD44v was closely related to the occurrence and development of many kinds of cancers, especially the selective expression of CD44v in metastatic tumors and its interaction with the cytoskeleton. As the CD44v had many complex forms of alternative splicing variants, a comprehensive comparison of homologous sequences of CD44v in NCBI was carried out from the structure, and the research progress and target therapy of CD44v in different tumors was comprehensively expounded.

    tFree radical plays a key role in the biological science. Under the normal situation, the equilibrium state of generation and elimination of the free radicals is assured to the health of the body. When the generation of free radicals increases and tissues fail to remove it, bio-molecules will be damaged. Many lesions are produced by the highly reactive hydroxyl radical and the DNA bases or the sugar moiety in the body. DNA can be permanently damaged by free radicals, manifested as decomposition of pentose and oxidation of bases. There is an obvious evidence for an important role of free radical-induced DNA damage in the etiology of numerous diseases. This paper provides an overview of the mechanisms of DNA damage, such as oxidation of bases, breakup of sugar moiety which induced by hydroxyl radicals. A deep understanding to the mechanisms of DNA damages induced by free radical would be of outmost importance for disease prevention and treatment, and the prospect of research in DNA damage by free radicals was also presented in this paper.

    Preparation and characterization of DEAE-agarose Langmuir-Blodgett films

    Diethylaminoethyl(DEAE)-Agarose was studied for preparing monolayers based on the Langmuir-Blodgett(LB) film technology. A high collapse pressure and a standard film forming process were found in the surface pressure (π)-area (A) isotherm of DEAE-Agarose. The effect of DEAE-Agarose concentration on the film formation showed that 0.5 mg/mL was the best. The surface potential increased with rise of the ligand density. DEAE-Agarose LB films were characterized with Brewster angle microscopy (BAM) and atomic force microscope (AFM) after being transferred onto silicon surface. The morphology of the film was pretty compact, well structured, and relatively smooth. The thickness was about 7.9 nm and the roughness was about 2 nm. This result indicated that the DEAE-Agarose LB film was successfully prepared and could be an novel material for the fabrication of LB films.

    Rapid preparation technology for tetrodotoxin based on immunochromatographic detection

    In order to establish a suitable pretreatment method for immunochromatographic detection for tetrodotoxin, in this study,the ultrafiltration technology was applied to optimize the pretreatment. The magnetic immunochromatographic test strip established by our laboratory was used for evaluation. The results showed that the optimal treatment of centrifugation at 4000 r/min for 10 min with a cut off molecular of 10 ku. This method was suitable for immunochromatographic assay with good recovery (92.4%-126.0%). Also, the concentration of protein in the solution reduced after pretreatment. This study provides technical support for rapid on-site test of tetrodotoxin.

    Optimization of extracting process of astaxanthin from Haematcoccus pluvialis

    This paper established a method for astaxanthin extracting by ultrasonic extraction and detection by high performance liquid chromatography(HPLC). The result showed that acetone was the optimal solvent among the selected solvents. The astaxanthin yield of Haematococcus pluvialis was used as measurement index, the sequence of extraction efficiency of different solvent was that acetone> methylene chloride: methanol(3∶ 1, V/V) > methanol > petroleum ether > chloroform. The mathematical model of ultrasound assisted astaxanthin extraction of H.pluvialis was established by response surface method. The optimal condition for extraction technique was that S/L 1∶ 377, ultrasonic time 5.19 min and ultrasonic power 145 W. Under the optimal condition, the predicted astaxanthin yield was 2.931%, and the measured value was 2.896%. It indicated that the adaptable of model was good. Compared with conventional reflux extraction methods, extraction efficiency of astaxanthin was improved significantly assisted by response surface method.

    The optimizition of exopolysacchrides production by an soil bacterium

    The soil bacterium L20309 with production capacity of extracellular polysaccharide was isolated from the prunus divaricata in Daxigou of Xinjiang and the fermentation conditions were optimized. Results showed that the better carbon and nitrogen sources for it to produce extracellular polysaccharide were glucose and peptone, and the optimal mass concentrations of carbon and nitrogen sources were 20 g/L and 30 g/L, respectively. The optimal growth conditions were pH 8.0 of the medium, the loading sample of 500 mL/L, the inoculum of 5% of 28℃. When cultured under such condition for 72 h, the polysaccharide product could reach to 2.562 μg/mL.

    tBilingual teaching in the experimental course of physiology is currently carried out. These efforts are aiming at improving bilingual teaching efficiency in the theoretical course of physiology and deepening the students′ understanding of related physiological knowledge. To achieve this, the curriculum design, educational mode, reference textbook, class hour arrangement, and after-class learning are all reformed. Evaluation mechanisms are also established to assess the effect of the above reforms. Moreover, based on the teaching feedbacks, we have deeply analyzed conundrums that restrict the development of bilingual teaching, including ambiguous teaching goals, lack of qualified teachers, and poor bases of the sophomore students. Possible solutions are also discussed to further improve and perfect bilingual teaching in the experimental course of physiology. So far, the reforms on bilingual teaching in the experimental course of physiology have greatly improved the students′ abilities in English application, clinical thinking, independent learning and dialectical thinking.

    CLARITY transparent brain technique is a cutting-edge experimental method in animal morphological investigation, which is lack of performing experiences in undergraduate student teaching. In this study, authors reconstructed the technical flow scheme to adapt undergraduate students according the current existing laboratory condition. By teaching practice, the students were trained in comprehensive studying ability and obtained advanced experimental skills.

    Smart phone and WeChat app has been very popular. So, to use smart phone as multimedia terminal device and to apply WeChat Official account platform as a means of communication will achieve instant messaging between teachers and students at any time and any place. Using of smart phone and WeChat app in the teaching of zoological field practice would be a nice approach for those teaching problems. Official platform will enable media courseware be pushed and make it convenient to answer questions of students; it will achieve broadcasting instant information of species distribution and give a guidance to students for their investigation and observation in field work.
