    Cloning and expression analysis of embryogenesis-associated protein EMB8 (DoEMB8) in Dendrobium officinale Kimura et Migo

    The embryogenesis-associated protein EMB8 (DoEMB8) in Dendrobium officinale Kimura et Migo was achieved by RACE and nest PCR technologies. The full length of DoEMB8 was 2077 bp, contained an open reading frame (ORF) of 1824 bp that encoded 608 amino acid residues. Phylogenetic analysis showed that DoEMB8 had the closest relationship with Phoenix dactylifera, Elaeis guineensis, Musa acuminata subsp. malaccensis and some other monocotyledon. DoEMB8 had 2 transmembrane helix structures and probably existed in mitochondrion analyzed by TMHMM and TargetP, respectively. RT-qPCR analysis showed that the expression level of DoEMB8 was relatively constant in different development stages of protocorm, while it was higher in PLB(Protocorm like body) compared to that in protocorm. In different tissues, the expression level of DoEMB8 was highest in seed and was 3.16 times higher than that in root, indicating that DoEMB8 had relatively constant and higher expression level in embryonic development and may play an important role in plant embryonic development.

    The objective of this study was to explore the correlation of JNK1 gene in the process of gonad development of Goldfish (Carassius auratus). The expression of JNK1 was studied and analyzed in different developmental phases of gonad by using quantitative real-time PCR and Western blotting. It was found that gonadosomatic index (GSI) of males and females showed significant seasonal variation with the development process of gonad and was the maximium during the breeding period (P <0.05). The qPCR analysis revealed that the expression of JNK1 firstly increased and then decreased at the development phases of ovary, reached the peak level at maturity phase (P <0.05),4.0 times higher before sexual maturity, 1.4 times higher degradation phase and 2.1 times higher at the rest phase compared with that at maturity phase.In the testis, the expression of JNK1 was found to be trace enhanced compared with before sexual maturity phase, however, the expression changes had no significant difference in the whole development process. Western blotting analysis showed that expression tendency of JNK1 protein was in line with that of JNK1 mRNA. Together, these results suggest that JNK1 plays an important role in the processes of the development of ovary and promoting the maturation of oocytes in fish.

    Morphological characteristics of antennal sensilla of female Sycophaga sp.observed with scanning electron miscroscopy

    Sycophaga sp. is a species-specific and galling parasite at the pre-female floral stage of Ficus auriculata. To provide a prerequisite for future electrophysiological studies on the fig wasp chemoreception, the antenna of female Sycophaga sp. was excised and examined by the sanning electron microscopy. The results showed that the antennae of Sycophaga sp. is geniculate in shape and consist of the basic segments: scape with radicula, pedicel and a long threadlike flagellum subdivided into 11 flagellomeres. The total antenal length is (1342.5±96.5)μm. Six sensillar types were recorded and described on the antennae of female Sycophaga sp.. The identified sensillar types included trichoid sensilla, chaetica sensilla, uniporous and multiporous basiconic sensilla, basiconic capitate peg sensilla, and multiporous placoid sensilla. Most sensilla were located on the outside laternal side and ventral side of the antennae. According to the abundance, trichoid sensilla and multiporous placoid sensilla were regarded as the main sensillar type on the antennae of the female wasps. The former was distributed over the whole area of antennal segments, except for the radicula, but the latter presented the equal number of sensilla on the 3rd-11th flagellomeres. Their probal functions were discussed in view of sensillar type, distribution and oviposition behavior of Sycophaga sp..

    The Yunnan shoot borers, Tomicus species, are indigenous shoot-feeding beetles that attack the Pinus yunnanensis in southern China. To provide a generally concentration on the physiological aspects and feeding divisions, the proventriculus of 4 Tomicus species, including T. yunnanensis, T. minor, T. piniperda and T. brevioplius, were analyzed with scanning electron microscopy. The results showed the adult proventriculus consisted of 8 chitinous plates are in lantern-like shape. Each plate is involved in the anterior plate and masticatory plate. The former has no median suture on the surface plate, which presents many transverse tooth lines with all types of denticles; while the latter is composed of declivity, stopping bristles and mastiutory brush. Together evolutionary relationship of proventricular structure corroborate the cuticular denticles and stopping bristles as well as substitute stopping bristles that play a critical role in feeding divisions among the scolytidae. Particularly, the spine-like hairs occurred on the median suture may indicate the niche difference of coadaptation on Tomicus species. The results will benefit for the further understanding the nutrient utilization and resistance ability of Tomicus species to the host trees.

    Effect of warm acclimation on body mass and energy metabolism in Eothenomys miletus

    In order to investigate the effect of warm acclimation on body mass and energy metabolism, parameters like body mass, body temperature, basal metabolic rate(BMR), non-shivering thermogenesis(NST), energy budgets and thermogenic activity of liver in E. miletus were measured. The results showed that warm acclimation increased body mass and body temperature, decreased energy intake and thermogenic activity, as well as decreased state Ⅲ of mitochondrial respiration of liver, but it did not affect state Ⅳ of mitochondrial respiration of liver. All of the results indicated that E. miletus under warm acclimation increased body mass and body temperature, reduced energy intake and heat production capacity as to adapt to high temperature stress.

    In order to study the effects of Pb2+ on tissue structure of liver, intestines and spleen of fish, Mudskippers (21.80±2.70)g were randomly stocked into 16 tanks with 8 fish each in triplicate. The fish were exposed to 0, 0.5, 1 and 2 mg/L Pb2+, and sampled on the 12th, 48th and 96th h. The results showed that Pb2+ exposure results in tissue edema, cytoplasm vacuolization and   in liver, epithelial cell damage, gobiet cells increased in intestinal, and lymphocytes increased in spleen. The results indicated that Pb2+ exposure can cause damage to the structure of liver, intestines and spleen of mudskipper.

    There are three different biological bait feeding modes to feed the Chinese mitten crab stage larvae, which are the individual feeding rotifers, rotifers and Artemia mixed feeding and the individual feeding Artemia. Parameters such as megalopae of metamorphosis to metamorphosis rate, metamorphosis megalopa of Megalopae survival rate and survival postlarvae (Megalopae and juvenile) weight after the experiment were used to discuss the investment and feeding modes of Chinese mitten crab. The results showed that among three biological bait feeding modes, the rates of zoea survival and metamorphosis at 96 h fed with the highest Artemia group were 33.33%±2.31% and 86.2%±7.2%, respectively. The average weights were not significantly different among the survival megalopa of the three different biological bait feeding modes. The average weight of survival of I larval was between (12.10±0.98)mg and (13.16±1.10)mg (P <0.05). The average length of survival megalopa fed with the Artemia was the best.

    Joint toxicity of bisphenol A and phenol to the juvenile prawn of Macrobrachium nipponense

    A static bioassay test was used to study the cute toxicity of BPA(Bisphenol A) and phenol to the juvenile prawn of Macrobrachium nipponens, and based on which, toxic unit (TU) method was used to study the joint toxicity of BPA and phenol to Macrobrachium nipponense. The results showed that there were significant relationships between the death rate of the juvenile prawn and the concentrations of BPA and phenol. The LC50 decreased gradually with the increasing of the exposure time, and there were significant dose-effect and and time-effect. The 24 h LC50 and 48 h LC50 of the juvenile prawn to BPA were 25.68, 12.03 mg/L, respectively, and 33.63, 27.59 mg/L to phenol, respectively. The safe concentrations of the juvenile prawn to BPA and phenol were 0.80 and 5.57 mg/L, respectively. The LC50 of 24 h and 48 h joint toxicity of BPA and phenol to the juvenile prawn were 0.97 and 0.51 TU, respectively. The TUi was less than 1, which meant that the joint action type is synergistic.

    In order to understand the tolerance of Mytilus coruscus to dry exposure in different temperature and humidity or dry conditions, we measured the juvenile with the shell length of (4.43±0.76)mm and (10.16±0.43)mm. The experiments were performed at low temperature (1℃,2℃, 4℃ and 8℃) and high temperature (30℃, 32℃ and 34℃) with the condition of being dry and moist, respectively. The results showed that the survive rate of juveniles after exposure and desiccation had a negative correlation with the time of the tolerance under the same humidity (P<0.05). And the semilethal time was 45-91 h at 1-8℃, while 17-23 h at 30-34℃. The endurance of juveniles to exposure and desiccation under low temperature was more than triple under high temperature. The ability to survival of large size of M. coruscus was stronger than that of the small size one. The semilethal time of small size juveniles was 70-79 h, while the semilethal time of lager size was 76-91 h at 4℃. The survival rate of M. coruscus under moisturizing conditions is higher than that in drying conditions (P<0.05). The survival rate about the group of rewet with slow change in temperature was higher than the quick change in temperature group after air expose in low temperature, but the result after high temperature exposure was the opposite. Research results that the optimal transportation condition of Mytilus coruscus was air exposured under moisture condition at 4℃. In this way, the survival rate could be guaranteed more than 90% in 24 h.

    tA comparative study on flesh composition including crude protein, crude fat, moisture, ash and 17 kinds of amino acids as well as the substance of inosine monophosphate (IMP) was conducted in Carassius auratus gibelio kept in fresh water and low salinity water (4‰ salinity), respectively. The results showed that carp kept in low salinity water had lower crude fat, moisture, ash content and higher crude protein content, and the contents were significantly different (P<0.05)except ash(cultivated in conpared with the contro fresh water). Amino acid and inosine monophosphate(IMP) contents were higher in Gibel carp kept in low salt water in fresh and dry(only detect IMP in fresh) muscle samples. The contents of Asp, Gly, Ala, Val, Leu and Phe in fresh samples and Asp, Ser, Gly, Ala, Tyr and Phe in dry samples were significantly different (P<0.05) while inosine monophosphate (IMP) in fresh and His in both fresh and dry samples were significantly different (P<0.01). The comparative analysis of essential amino acids, delicious amino acids, sweet amino acids content in fresh and dry samples indicated that the contents were significantly different except delicious amino acids in fresh samples. The contents of delicious amino acids, sweet amino acids in dry samples were significantly different (P<0.01).

    Through changing the plant growth regulators proportion of medium, the optimal medium for callus proliferation and high content of escin were obtained; the growth of callus and accumulation of secondary metabolites were preliminarily analyzed which can provide theoretical basis for the production of escin in cell culture. Results revealed that the optimal medium for callus proliferation of embryos in Aesculus hippocastanum L. was MS+8 mg/L 6-BA+1 mg/L NAA. In the optimal medium, the best subculture period was 35 d and the best harvest time was 42 d. The callus specific growth rate and escin specific synthesis rate were analyzed, which showed that with the decrease of the callus growth rate and specific growth rate, the escin synthesis rate and specific synthesis rate was increased.

    A study on flora of the national key protected wild plants in East China

    Based on field investigation and data access, this paper made a the list of national key protected wild plants and analyzed the species composition and flora of the national key protected wild plants in East China. Results showed that there were 94 species of national key protected wild plants belonged to 69 genera and 42 families, among which there were 60 species of angiosperms belonged to 49 genera and 30 families, 22 species of gymnosperms belonged to 14 genera and 7 families, and 12 species of ferns belonged to 6 genera and 5 families. The origin of national key protected wild plants was ancient and the composition was complex, and endemic in this area was obvious. Tropical and temperate elements were dominant in the flora, which took up an obvious characteristic of transition. The analysis of flora richness and similarity indicated that the species in Jiangxi, Zhejiang, Fujian and Anhui were relatively abundant and closely related to each other, and that the species in Jiangsu and Shanghai were scarce, and meanwhile they were less related to the other provinces. The species of national key protected wild plants in Shandong was least, without the same families, genera, species as other provinces. The aim of the research was to analyze flora of the national key protected wild plants, in order to provide references for reasonable conservation and effectively use of the key protection wild plants in East China.

    Gap, caused by natural disturbance, can cause the change of light and temperature in the forest patch obviously. Based on an investigation in a sample of a Pinus dabeshanensis forest in Dabieshan Mountains, we recognized three patch types including gap, gap adjacent and closed canopy and analyzed their microclimate characteristics. The daily change patterns of microclimate characteristics in three patch types were different. The light intensity and air temperature were much greater in gap than in the gap adjacent and closed canopy. The light intensity, air temperature and soil surface temperature had significant difference under different patch types.

    Susong Huayang Lake, as the typical linking lakes of the Yangtze River, is one form of the ecological barrier in the eastern China. It has a surface area of 183 hm2. In order to strengthen wetland protection and restoration of aquatic vegetation, from March 2015, three types of ecological reconstruction experiments have been carried out. Main applied measurements include cultivating Zizania latifolia seeding, Nuphar pumilum seeding, Nelumbo nucifera rhizome, broadcasting Trapa quadrispinosa fruit, Vallisneria natans fruit, Euryale ferox seed and so on. The results showed that the seeding of Zizania latifolia and Nuphar pumilum can grow well with 14 aquatic macrophyte association being set up. The coverage of Ass. Zizania latifolia and Ass. Nelumbo nucifera are 90% and 95%, respectively, while the biomass of Ass. Zizania latifolia and Ass. Nelumbo nucifera are 10.57 kg/m2 and 2.05 kg/m2, respectively. All the Trapa quadrispinosa fruits, Vallisneria natans fruits and Euryale ferox seed can germinate early and form seedling, but gradually disappear affected by the fishery. According to the result of this study, it is important to rehabilitate Ass. Zizania latifolia and Ass. Nelumbo nucifera in littoral of lakes, and to facilitate the grow of submerged plant communities and floating plant communities by controlling the fishery breeding intensity, hydrological conditions, and establishing submerged macrophytes and floating macrophytes in the lake.

    Dynamics of the Vibrio community in aquaculture area of Xiangshan harbour, Ningbo

    The Xiangshan harbour is a typical aquaculture area in China. Vibrio is one of the most important pathogenic bacteria, which is the main cause of bacterial disease of marine fishes. In this study, seawater sample and fish (Pseudosciaena crocea) in the Xiangshan harbour aquaculture area were collected from Aug. to Nov., 2012. The number of Vibrio and total bacteria and the environmental parameters were monitored. Vibrio community structure changed significantly with time. Canonical correspondence analysis showed that temperature, TOC, PO3-4-P and NO-3-N were the most important environmental driver factors of the Vibrio community structure. The predominant bacteria of seawater sample and the fish were identified using 16S rDNA gene and rpoA gene. The result demonstrated that there was obvious difference between the cage-area and control area of Vibrio species component. The abundance of Vibrio harveyi increased suddenly when the fish disease outbreak occurred. Hence, the abundance of Vibrio harveyi can be an indicator of Xiangshan harbour fish illness. Moreover, Photobacterium angustum increased sharply as a result of antibiotic application after the outbreak.

    Endogenous microbial diversity in Tarim oilfield based on deep well in-situ sampling technique

    To insight into the endogenous microbial communities and potential functions living in high pressure and high temperature (HPHT) reservoirs of Tarim Oilfield, crude oil pressure-volume-temperature (PVT) properties equipment was operated for deep wells in-situ sampling in both LN-A and TZ-B wells selected in Lunnan and Tazhong zone, respectively. Meanwhile, wellhead samples were collected as the control. The differences of microbial community diversity in oil-water samples were compared and analyzed. The metagenomics results showed that the dominant microorganism from in situ samples of LN-A well was Bacillus (56%), which of TZ-B well was Pseudomonas (50%). Comparing with samples from wellhead, the dominant microorganism was in accord with that of in-situ, however, the percentage was a little bit higher. The results of the two sampling methods showed that the microbial community structure was richer, dominant bacteria was dispersed, and more new functional microorganisms were found in deep well group (in-situ and wellhead samples were 14 and 3 species from LN-A well, respectively, while those of TZ-B well were 12 and 3 species). In this study, deep well in-situ collection, metagenomics and microbial community analysis technologies were integrated, which can help us fully understand and probe in-depth the functional microbial resource in HPHT reservoir. Additionally, this article can provide practical instructions and application value in microbial enhanced oil recovery technology performed in HPHT reservoir.

    DREB/CBF (Dehydration responsive element binding protein/C-repeat binding factor)transcription factor, the drought responsive element-binding protein/C-repeat sequence,can specifically interact with the DRE/CRT (Dehydration-responsive element/Cis-acting elements). It activates the expressions of genes associated with abiotic stress resistance in plant cells and thus enhances the plant resistance to various stresses. In this paper, the structure, function, species distribution and signal transduction of DREB/CBF transcription factors were systematically introduced. The roles and existing problems of DREB/CBF transcription factors in plant genetic engineering breeding were discussed.All these above are aimed at providing references for the further research and application of DREB/CBF transcription factor.

    Cadmium has strong biological toxicity. It could be absorbed by smoking, dietary and inhalation. Cadmium could lead to series damage to mammalian, including bone misshapen, kidney injury and cancer; it can also lead to abnormal lipid metabolism. In recent years, many researchers found that cadmium could lead to cardiovescular diseases in mammalians, such as heart attack, hypertension and so on,however, the mechanism is still unknown. The mechanism of this phenomenon could be the result of ROS(Reactive oxygen species) production, Na+/K+ ATPase activity inhibition and impaired VE-cadherin function. Here, we discussed the research progress in mechanisms of cadmium induced hypertension.

    tBiological network is a kind of typical complex network, thus the methods for identifying of the key nodes in biological networks are mainly from general complex network methods. This paper introduces several methods for the key indicators in the general complex networks, and then sums up the methods for the identification of key nodes in biological networks. It points out the differences between the biological network key nodes recognition and other general complex networks. The research directions is also discussed.

    Optimization of immature embryo rescue technique system for persimmon

    The high percentage of abnormal seeds during the growth of its hybrid seriously hampered the development of the persimmon cross-breeding research. The technique of immature embryo rescue could rescue abortive embryos to obtain hybrid seedlings. The immature embryo of D. kaki Thunb ‘Fuyu’, D. kaki Thunb ‘Okugosho’, D. kaki Thunb ‘Taishu’ was used as the experimental materials, an orthogonal experiment of L9(34 )was adopted to study for embryo rescue. Then, using the immature embryo of D. kaki Thunb ‘Taishu’as the experimental materials, media and embryo-collecting time were respectively determined as univariate factor for further single factor experiment. The results showed that according to the effect on immature embryo rescue,the order for the factors was as follows: media, embryo-collecting time, persimmon varieties, and cultivation conditions. The seedling rates of embryos from 45 days after pollination (DAP) to 55 DAP were extremely low. Though, the embryo culture seedling rates increase as embryo age increased. The seedling rates of embryo from 77 DAP dropped slightly. So, the optimal embryo-collecting time for persimmon immature embryo rescue in the lower area of the Yangtze River is: 62-69 DAP. An extensive variation existed in the development process, embryo seedlings rate, seedling height and root number of persimmon immature embryo in different medium. By comparative analysis, the optimal medium for persimmon immature embryo rescue is 1/2 MS+ZT (0.4 mg/L) + IBA(0.2 mg/L)+Activated carbon (3 g/L)+Sucrose (70 g/L). After cultivating for 45 days, the average seedlings rate, average seedling height and average root number of persimmon immature embryo can, be up to 0.88, 3.52 cm and 2.52 respectively.

    Nine mycorrhizal fungi were isolated from the roots of Vaccinium Ashei in southern Anhui Province. The inoculation experiments for the mycorrhizal fungi were performed to detect the survival rate and the height in the shoots and to test the nutrient elements and chlorophyll content in plants of blueberry. four Fungi strains were found as dominant strains in promoting growth of Vaccinium Ashei. Through morphological observation and molecular identification method, we found that three strains belong to families Mucor species Mucor circinelloides, and the different among them may be in subspecies level. Another species belongs to families Mucor species Mucor racemosus, and the similarity between them was greater than 99%. The mycorrhizal fungi from families Mucor species Mucor circinelloides and Mucor racemosus can significant promote the nutrient absorption and growth of Vaccinium Ashei.

    In order to improve the teaching quality and enhance the students′ listening interest and cultivate the creative talents, this paper designs the exploration and reform of the teaching methods for the insect taxonomy in the applied undergraduate colleges by enhancing the application of the knowledge of insect taxonomy, including the setting of the teaching method of the taxonomy of insects, the establishment of the target, the way of interaction and discussion. The benefits of student participation, role transformation, and group discussion on the students′ grasp of insect taxonomy were analyzed. The interaction between teachers and students in classroom teaching and group discussionis was an organic unity of teaching and learning, which not only is the requirement of teaching reform, but also is the inevitable choice to improve the teaching effect.

    In order to improve the quality of the higher vocational education, we make the pertinence and appropriate training plan for the students of five-year vocational college. This article gives the comparison evaluation on the effects of non-directive teaching pattern for the five-year vocational students, and the result showed that with the non-directive teaching pattern both teacher-student relationship and learning interests were improved, meanwhile, the mastery of knowledge was better than that with the traditional teaching model.
