    Differential expression of N-glycans in hypoxia-induced human malignant breast tumor cells MDA-MB-231

    Tumor hypoxia is a common pathological feature in many cancers. Hypoxic microenvironments dramatically shift the pattern of intracellular glucose metabolism, which increase glucose uptake and nucleotide sugar levels. In this study, hypoxia-induced malignant breast tumor (MDA-MB-231) cell model was established. Under hypoxia conditions, MDA-MB-231 cells showed the fibrosis morphology. Expression level of β-catenin in hypoxia-treated cells was reduced, fibronectin was increased, and cell motility was enhanced, which indicated that hypoxia induces the epithelial-mesenchymal transition. The structures of N-glycans from normoxia and hypoxia-treated cells were further analyzed by matrix-assisted laser desorption ionization time of flight mass spectrometry (MALDI-TOF-MS). Seventeen N-glycans were differentially expressed in normoxia and hypoxia-treated MDA-MB-231 cells. The high-mannose and tetra-antennary type of N-glycans were increased, while the hybrid type, bisecting type, biantennary type, triantennary type, fucosylation and sialylation of N-glycans were decreased in hypoxia-treated MDA-MB-231 cells. The study of lectin Con A and LCA fluorescence staining consistented with MS analysis. The present research indicates that hypoxia alters the expression of N-glycans in malignant breast tumor cells, which may provide the fundamental observations for further understanding functional roles of differential expression of N-glycans in hypoxia-treated cells.

    Analysis of N-linked glycans in bladder cancer

    Bladder cancer is one of the most common cancers and which occurs in the bladder mucosa.The aberrant changes of glycans have been shown to be associated with tumorigenesis, tumor progression and metastasis. The study focused on structures and functions of tumor-specific glycans was useful for the discovery of new candidate biomarkers of bladder cancer. In this study, N-glycans in glycoproteins in 20 bladder cancer and 11 normal samples were released by PNGase F, and sialic acids were modified by acetohydrazide. A total of 42 N-glycan structures were identified and 15 N-glycans were thought to be candidate biomarkers. It indicated that complex type and sialylation type N-glycans were increased significantly, while the high-mannose type and bisecting N-glycans were decreased significantly in bladder cancer sera. These findings showed the expression of glycan structure is associated with the occurrence of bladder cancer, which is important to reveal the glycan mechanism in the development of bladder cancer. The differential glycans could be useful for the early diagnosis.

    Characterization of xanthan gum produced from glycerol by Xanthomonas campestris CCTCC M2015714

    This study focused on the xanthan gum produced by Xanthomonas campestris CCTCC M2015714 with carbon source of glycerol. The monosaccharide composition of xanthan gum was glucose, mannose and glucuronic acid, the molar ratio of three monosaccharides was glucose∶ mannose∶ glucuronic acid=2.0∶ 1.65∶ 1.0. The results of FT-IR and NMR showed that xanthan gum produced from glycerol had similar functional groups and chemical structure comparing with commercial xanthan. The molecular weight of xanthan gum produced using our method was 3.0×106 u, which was about half of the commercial xanthan (6.4×106 u). Moreover, the viscosity of xanthan gum produced from glycerol was significantly lower than that of the commercial xanthan. Depending on this work, xanthan gum produced from glycerol exhibited great characteristics and also supplied a valuable method for glycerol application.

    Effects of glycosylated modification on emulsifying properties of collagen

    In order to improve the stability and functional properties of collagen, a glycosylated product of collagen was prepared by Maillard reaction. The result of fluorescence spectra indicated that the Maillard reaction between the collagen and lactose occurred successfully. The fluorescence intensity was stronger with the longer reaction time. The studies on the reaction conditions showed that the reaction rate and the browning index of the products were increased with the increase of reaction time, temperature, pH and concentration of lactose. The emulsifying property of collagen was improved while the average micelle size in solutions increased after the glycosylation.

    The objective of this study was to construct eukaryotic expression vector of human interleukin-7, and make it expressed in HEK293 cells. IL-7 gene was amplified by PCR from the lentivirus expression vector pLV-CMV-EF1-GPL-rIL-7, and then cloned into the pcDNA3.1 vector to construct eukaryotic expression vector pcDNA3.1-rIL-7. The constructed plasmid was transfected into the HEK293 cells via LipofectamineTM 2000 separately and screened by G418 continuously. By G418 screening and monoclonal, stably secreting expression IL-7 cell line HEK293-rIL-7 was obtained, successfully constructed eukaryotic expression vector pcDNA3.1-rIL-7, and expressed in HEK293 cells. By cell cloning, one cell line with high secretory expression was obtained, named HEK293-rIL7-2G3. The content of IL-7 in the supernatant after 48 h was 3.2 ng/mL.

    FoxP2 is the first gene involved in human speech. This gene also exists in birds and plays pivotal roles in development, adulthood, and avian vocal learning. The sequences of FoxP2 in five avian species and their related species were retrieved from GenBank and Ensembl. The sequence characterization, evolutionary rate of FoxP2 were analyzed. The phylogenetic analyses of five avian species and two reptile species, with two mammalian species as outgroup, were conducted. Using maximum parsimony (MP),maximum likelihood (ML), and Bayesian inference (BI) methods based on the nucleotide sequence of FoxP2, the monophyly of five avian species was confirmed. Our results may provide useful information about the evolution of FoxP subfamily in birds.

    An experiment was conducted to study the individual or joint toxicity of aflatoxin B1 (AFB1) and deoxynivalenol (DON) in mice. Ninety-six Kunming male mice (21-day-old) were randomly divided into four groups, three replicates per group and eight per replicate. All mice were fed with the same feedstuff during the experiment. Mice in group Ⅰ were gavaged with the physiological saline every day; the mice in group Ⅱ were gavaged with 200 μg·kg-1 AFB1; the mice in group Ⅲ were gavaged with 500 μg·kg-1 DON and the mice in group Ⅳ were gavaged with 200 μg·kg-1 AFB1 + 500 μg·kg-1.The trial was conducted for 30 days. On the 15th and 30th day, four mice from each replicate were selected randomly and blood samples were taken from the eyeball for measuring serum biochemical indexes. Meanwhile, the selected mice were slaughtered and the liver and spleen were taken for calculating the organ relative indexes, and the pathologic changes of liver tissues were observed. The results showed that, compared with the control group, the relative organ indexes of liver and spleen, serum AST activities and MDA contents of mice in Group Ⅳ were significantly increased , the ALB and TP contents were significantly decreased on the 15th day. Compared with Group Ⅱ and Group Ⅲ, the relative organ indexes of liver and spleen, serum AST activities and MDA contents of mice in Group Ⅳ were significantly increased. On the 30th day, compared with the control group, weight gain of mice in Group Ⅱ, Ⅲ, and Ⅳ were significantly decreased; serum ALT, ALP, r-GT activities of mice in Group Ⅳ were significantly increased, but SOD activities, and TP, ALB contents were significantly decreased. Compared with Group Ⅱ and Group Ⅲ, serum ALP and r-GT activities, and MDA contents of mice in Group Ⅳ were significantly increased, while serum SOD activities were significantly decreased. These experimental results suggest that AFB1 and DON can decrease the weight gain, liver and spleen relative indexes, and alter the biochemical indexes; the joint toxic effects of AFB1and DON are higher than any of the individual toxicology.

    Suppression of melanin formation by floridoside

    To study the inhibitory effect of floridoside in red algae on the generation of melanin, the B16-F0 melanoma cells were used to evaluate the ability of floridoside on the biosynthesis of melanin. The effect of floridoside on the activity of tyrosinase was also detected for investigating the mechanism. Results indicated that floridoside showed no obvious toxicity to B16-F0 melanoma cells, but suppressed melanogenesis with the concentration dependence manner. Floridoside was discovered to inhibit both the monophenolase and the o-diphenolase activities. It influenced the steady state of monophenolase activity and IC50 was 89 mmol/L. The inhibition on o-diphenolase activity was in a concentration dependent manner, and 40 mmol/L floridoside reduced the steady state activity for 33.08%. The inhibition kinetics analyzed by Lineweaver-Burk plots showed that floridoside was a reversible competitive inhibitor, and the inhibition constant was determined to be 16.44 mmol/L for the free enzyme. In general, floridoside was potential to suppress the generation of melanin, and the mechanism was to inhibit the activity of tyrosinase, especially the activity of o-diphenolase.

    Pharmacokinetics and residues of allicin in crucian carp

    The pharmacokinetics and residues of allicin were studied in crucian carp exposed to oral administration at 7.5 mg/kg bodyweight by means of gas chromatography (GC).The results showed that the concentrations versus time of allicin in muscle was well described by two-department open model with first-order absorption after a single oral administration.The residues in crucian carp by oral administration for 3 d once a day were found to be less than the maximum residue limit (MRL) (10 μg/kg) at the day 13 for DADS and the day 15 for DATS. The findings indicated that the withdrawal time should be 15 days after administration on these conditions.

    Using the histological methods, the gonadal development, structures of gonad, spawning time and type of reproduction in Tribolodon brandti were studied. The testes of T.brandti have typical lobular structures, and can be divided into six developmental stages. According to the characteristics of the ovaries and the types of ovarian germ cells, the oogenesis process can also be divided into six developmental stages. The microscopic observation showed that the gonadal development of sexual mature T. brandti differentiated to stage III at the end of September, and stayed at the phase III in the winter period. While the gonadal development of immature juveniles age 2+ was at the phase Ⅱ and remained such stage during the winter period. In the late April and early May, gonadal development of sexual mature males differentiated to the phase V, but in the females it was until late May or early June that they reached to the developmental phase V. Taken together, gonadal development of the males was faster than that of the females. No significant difference was observed in the gonad development of 2-3-year-old juvenile cultured in either fresh water or weak alkali water environment. The body weight 2+-year-old group in the weak alkali water were significantly higher than that in the fresh water, but no significant difference was found between 3+-year-old groups reared in weak alkali environment and those in fresh water. Meanwhile, significant differences of body weight existed between females and males in the weak alkali as well as in the fresh water. In the gonad development cycle, the average condition factor of female and male ranged from 1.313 to 1.697. During the sexual cycle, the male gonad index began to rise significantly from the II period, while the female gonad index was significantly different among development periods, with the peak value of 14.22 in the period of V. The conclusion could be drawn that the gonadal development peak of T. brandti appears only once a year, the ovary is partial synchronic, and this species is iteroparous.

    Differences of fatness and nutrient content in artificially and naturally ripened Mytilus coruscus

    To study the differences of fatness and nutrient content of gonad between naturally and artificially ripened Mytilus coruscus, fatness and nutrient contents in three developmental periods were sampled and analyzed. The results showed that the fatness changed consistently with the content of crude protein and crude fat, and achieved the peak in mature period. The fatness was 18.80% and the crude protein and crude fat was 62.57% and 21.05%, respectively, in the artificially ripened Mytilus coruscus, which were higher than those in the natural ripened shells. The contents of mineral substance in growing period and mature period of natural ripened Mytilus coruscus were 32.18% and 18.27%, respectively, which were comparatively higher than those in the artificially ripened Mytilus coruscus. The moisture content showed little difference between the two ways of maturity. These findings suggested that the artificially ripened Mytilus coruscus showed obvious advantages in fastness and moisture content.

    In order to investigate the respiratory metabolism of Portunus trituberculatus, the closed hydrostatic method was used to determine the oxygen consumption rate, ammonia excretion rate and asphyxiation point at the growth period of Ⅵ-Ⅺ. The results showed that the oxygen consumption increased with the late growth periods, the maximum value was 3.950 mg/L at Ⅺ period, the differences were significant among Ⅷ-Ⅺ period (P<0.01); the oxygen consumption rate decreased with the large growth period. The ammonia excretion and ammonia excretion rate increased between Ⅵ-Ⅹ period, and the maximum values were 1.34 mg/L and 0.017 mg/(g.h), respectilely at Ⅹ period. The asphyxiation point increased between Ⅵ-Ⅹ period, and the maximum value was 9.12 mg/L at Ⅹ period.

    Pteroceltis tatarinowii Maxim., the main raw material for manufacturing of rice paper, has a high economic value. In order to obtain high-quality bark of P. tatarinowii and provide theoretical guidance for artificial cultivation of it, this paper selected six provinces around Anhui Province where abundant P. tatarinowii distribute, and compared fiber length, width and length-width ratio of P. tatarinowii there. Results showed that the fiber length, width and length-width ratio of P. tatarinowii varied with its growing environments and has a significant difference. The P. tatarinowii in Qingtansi, Shandong Province and Jingxian, Anhui Province all has high quality and its length-width ratio is 283.8 and 229.55, respectively, which were obviously higher than that in other four places; Correlation analysis showed that the quality of P. tatarinowii bark has a certain degree of positive and negative correlation with its various ecological factors; the principal component analysis indicated that the accumulated contribution rate of three principal components reached 94.66%, and the rank of each factor for the accumulated contribution rate from better to less was: temperature-light factor, soil factor and location factor, which manifested that the temperature-light factor had the largest effect on the quality of P. tatarinowii.

    The contents of the total polysaccharide and the 8 trace elements of 1-7 years-old Bozhou Peony were detected by anthracenone-concentrated-sulfuric-acid colorimetry and AAS, respectively, to explore the dynamic accumulation of polysaccharides and 8 trace elements in Bozhou Peony Paeonia lactiflora, and to provide scientific basis for the optimum harvesting age, quality evaluation and standardized cultivation of Bozhou Peony. The results indicated that cultivating ages would significantly impact the accumulation of polysaccharide and trace elements. The average contents of polysaccharides of Bozhou Peony (from 1 to 7 years) were:(2.06±0.12)%, (3.59±0.17)%, (4.66±0.26)%, (3.14±0.19)%, (2.44±0.03)%, (2.49±0.12)% and (2.94±0.20)%; the mass fractions of Cu, Pb, Cr ,Mg, Ca, Na, Zn and Ba were (4.14±0.39), (5.02±0.87), (1.69±0.07), (300.17±13.64), (1476.66±57.49), (71.88±2.26), (8.58±0.56) and (369.51±26.68) mg/kg(DW), respectively, and all the mental elements were in limits except Pd content in the samples exceeded the standard limit 0.89 mg kg(DW). Our experimental suggested that the Bozhou Peony cultivated for 3 years obtained the highest polysaccharide. There were rich essential mental elements in Bozhou Peony and the contents of heavy mental elements were lower than the limited. There might exist correlation between the trace elements contents and the accumulation of polysaccharide in Bozhou Peony.

    According to the forest cycle theory, forest communities are the embedded systems with different development patch types. The heterogeneity of the photosynthetic characteristics of Pinus dabeshanensis seedling in different patch types (gap patch, building patch, mature patch and degenerate patch) were studied. Our results were as follows: compared to other patch phases, in gap patch phase, the seedlings of P. dabeshanensis exhibited the highest net photosynthetic rate (Pn), stomatal conductance (Gs), transpiration rate (Tr), carboxylation efficiency (CE), and the maximum quantum efficiency of PSⅡ (Fv/Fm). The diurnal variations of coefficient of photochemical quenching (qP), coefficient of non-photochemical quenching (qN), PSⅡ quantum yields (ΦPSⅡ) and electron transport were (ETR) the highest in the gap patch, and the lowest in the mature patch. Our results suggest that the gap patch is the optimal environment for their growth, and the mature patch inhabits the growth of P. dabeshanensis seedling.

    Biological motion processing in individuals with high autistic traits

    To explore biological motion perception in individuals with autistic traits, we screened 35 individuals with high autistic traits by AQ as autistic traits (AT) group. Another 35 individuals with low autistic traits were included as controls. All subjects took the time lag judgment task to test their abilities of biological motion perception. As results, in biological motion task with social cues, there was a significant difference between the control group and AT group (-0.095±0.336 vs 0.104±0.252, t=-2.796, P<0.05) regarding the PSE values. The PSE value of the AT group was positive. Compared with "0", the difference was statistically significant (t=2.434, P<0.05), which means a "time distance" compression effect. In biological motion task without social cues, there was no significant difference between control group and AT group (-0.079±0.250 vs -0.083±0.306, t=0.061, P=0.951) regarding the PSE values. In conclusion, the individuals with high AT have deficit in perceiving biological motion with social cues. These results implicate that autistic traits can affect the ability to perceive biological motion.

    microRNA (miRNA) is a class of non-coding RNA, which usually regulates target genes by bonding with the 3′noncoding region (3′UTR) of mRNA and forms an induced silence complex. There is an intimate connection between miR-155 and immunity. Also, miR-155 plays irreplaceable role in differentiation and function of immune cells, including dendritic cells, macrophages, B cells and T cell. There are collected 25 kinds of miR-155 species in miRBase datebase, ranging from coelenterate to human being and other higher animals. Among them there are six kinds of aquatic animals, including the Atlantic salmon, zebrafish, spot fork tail madtoms, catfish, and brocade carp, etc. For providing references for the study of the function of miR-155, we reviewed the immune function of miR-155, the progress of its target genes and the researches in aquatic animals.

    The research development of lncRNA in esophageal squamous cell carcinoma

    Esophageal squamous cell carcinoma(ESCC)is one of the most fatal malignant tumors in digestion system. Long non-coding RNA(LncRNA)has become a research hotspot since it is not only involved in gene expression and regulation in different levels, but also affects cell proliferation, differentiation, metabolism and apoptosis. In this review, we first briefly introduce the structure of lncRNA and then summarize the recent research progresses of lncRNAs in the field of ESCC. LncRNAs regulate gene expression of esophageal squamous carcinoma associated genes by epigenetic modification, alternative splicing and co-expression network,furthermore, affect the development and progression of esophageal squamous cell carcinomas. Therefore, LncRNAs may serve as markers for ESCC diagnosis and prognosis and even serve as therapeutic targets. Secondary structure prediction of lncRNA is helpful to promote the research of lncRNA.

At the same time, some species can carry pathogens that cause allergic reactions such as rhinitis and asthma. Mongolia is located in the east part of the Palaearctic Region, its classification research foundation is poor and no taxonomic study of the animal. After nearly 40 years of research and development and according to complete statistic, Mongolian Chironomidae has recorded 269 species, belonging to 6 subfamilies. This paper made a review on the history and recent progress in the study of Mongolian chironomidae and provided the latest list in order to provide data for taxonomic research

    Assessment of a high throughout detection method of antibody concentration by bio-layer interference technique

    This research focused on the high throughout method for developing of antibody concentration detection based on the Bio-layer interferometry (BLI). The method gained the optimum efficiency with a 2 minutes binding on protein A sensor at a temperature of 30℃ and a shake speed of 400 r/min. The validation includes specificity, repeatability, accuracy and durability. The results showed that it fit a good linear relationship within an antibody concentration range of 0.781-400 μg/mL; relative deviations of repeatability including precision, intermediate precision and inter-day precision were below 2.5%. The accuracy ranged from 94.46% to 114.00%, the detection limit was 0.781 μg/mL and the limit of quantification was 1 μg/mL. The results certified the reliability of the method. The method can be used for detection of samples with a large scale benefitt from its high speed and throughout.

    The development of chemiluminescent immunoassay detection kit for 25-hydroxy vitamin D

    In this paper, a chemiluminescent immunoassay (CLIA) kit for quantitative assay of 25-hydroxyvitamin D(25-OHD)in serum was developed and evaluated. The kit is established as follows: first, 25-hydroxyvitamin D monoclonal antibodies was coated with the 96 well microplate, then the serum sample and the release reagent were added into the microplate. After incubation, the 25-OHD of the sample was fully released and combined with the 25-OHD antibody coated on the microplate. HRP conjugate (mixture of biotinylated 25-OHD and streptavidin-labeled HRP ) compete with the 25-OHD released from sample to antibodies. After washing and adding the CLLIA substrate, the relative luminous intensity was read with chemiluminescent immunoassay analyzer. The minimum detection limit of the kit is 4.9 ng.mL-1, with the bias of less than 10%. The intra-assay and inter-assay coefficients of variation were less than 10%, and 15%, respectively. Its preserving duration could be as long as 12 months at 2-8℃. Two hundred clinical serum specimens were tested with this kit and a foreign made serum ferritin kit, the correlation coefficient was 0.9717. 25-hydroxy vitamin D (CLIA) quantification detection kit was successfully prepared, and all the technical parameters reached the industrial standards. The developed kit is sensitive, stable, fast and simple.

    Natural selection plays an important role in changing the gene frequency of a population. The change of population genetic structure caused by the selection is an adaptive change. Therefore, natural selection is considered to be the main factor of species adaptive evolution. Considering the importance of the concept in explaining biological evolution, it has received much attention in the teaching of genetics. In order to better display the effect of natural selection on genetic equilibrium, Excel 2003 is used to simulate the selection process, and it is expected to understand the selection process more vividly and profoundly.

    It is important and also difficult for college students in such majors as veterinary science to study knowledge of animal virus taxonomy. Finding a suitable method to learn classification of a great number of veterinary viruses with abstract concept and their current taxonomic status as soon as possible plays a simply leading role in understanding structure, proliferative characteristic, pathogenicity, antigenicity and variability of a particular virus and conducting viral isolation and culture, detection and identification, vaccine development and disease control. The author made out a set of formulas in rhyme covering conventional veterinary viruses and found it useful for students to realize rapid memorization in the teaching trials.

     Animal disease diagnosis and treatment course practice is a new course offered to students majoring in veterinary medicine in our university. There is no existing method available for reference.The practice mainly include diagnosis and treatment of ruminal acidosis model in goats. The teaching method, the principle of student as main position and teachers as secondary position is adhered to. Many procedures of animal disease diagnosis and treatment course practice are explored and improved constantly. The modeling and diagnosis and treatment method suitable for located goats are explored. Reasonable assessment program is drew up. The results from questionnaire indicate that the teaching method are supported and affirmated sincerely by students. However, mature teaching methods and implementation program are explored in the process of animal disease diagnosis and treatment course practice, further exploration and improvement are still needed.

    In order to improve the teaching effect of the course of "medicinal botany", this paper tries to apply the semi-open teaching model into the teaching practice of this course, taking the chapter of "organs of plants" as an example. In the semi-open model, teachers guide students to learn and consolidate basic concepts describing the characteristics of plant organs by themselves, using the plant materials around. Application of the new teaching mode boosted the enthusiasm of the students‘learning, trained students’ ability to think independently and gain knowledge, and improved the students′ learning achievement. Students were actively willing to participate in the autonomous learning under the semi-open mode, and hope to extend this model into more practice of courses teaching.
