Abstract: DNA methylation is an important epigenetic mark in many eukaryotes. The quantitative measurement of mammal′s genome-wide DNA methylation level has become a research hotspot by combining with the next-generation sequencing (NGS) technologies. With the advantages of high-throughput, high coverage and high resolution through the comparison with other sequencings, NGS technologies provide an important approach for accurately detecting DNA methylation sites within the whole genome. In order to provide a reference for the selection of DNA methylation sequencing technology, three methods of restriction endonuclease digestion, affinity enrichment and bisulfite conversion were summarized and compared in this paper, and the future development of sequencing technology was predicted.
