前额叶皮质GABA能中间神经元在抑郁症发病及治疗中的作用

The role of prefrontal cortical GABAergic interneurons in the pathogenesis and treatment of depression

摘要: 综述抑郁状态下前额叶皮层(Prefrontal cortex, PFC)形态、功能的改变和γ-氨基丁酸(γ-aminobutyric acid, GABA)能中间神经元及其亚型数量与生理功能的变化。探究不同种类的抑制性神经元在抑郁症发病过程中的形态及电生理特性等微观变化的影响,并通过抑制性神经元的生理、病理改变进而对锥体神经元产生去抑制,最终影响抑郁症的发展进程进行阐述。通过比对临床前和临床中抗抑郁药物的疗效,关注抗抑郁药如何通过改变GABA能中间神经元的功能及表达水平从而进一步改善抑郁样状态,主要讨论GABAA受体和GABAB受体作为治疗靶点参与抗抑郁药治疗作用机制。最后,剖析PFC中GABA能中间神经元参与抑郁症发病的现存问题,展望以GABA能中间神经元为中心,探索新的受体、靶点、通路的未来研究方向,以期为抑郁症相关研究及其临床治疗提供参考。

Abstract: This article reviewed the changes in morphology and function of the prefrontal cortex (PFC) and the alterations in the number and physiological function of GABAergic interneurons and their subtypes in depression. The impact of microstructural changes in different types of inhibitory neurons on the pathogenesis of depression, including their morphology and electrophysiological characteristics, was explored. The physiological and pathological changes in inhibitory neurons were discussed to elucidate their role in disinhibition of pyramidal neurons and the subsequent development of depression. The effects of antidepressant medications on GABAergic interneurons, including changes in their function and expression levels, were compared between preclinical and clinical studies. The involvement of GABAAand GABABreceptors as therapeutic targets in the treatment of depression was emphasized. Finally, the existing problems related to the involvement of GABAergic interneurons in the pathogenesis of depression in PFC were analyzed, and future research directions focusing on new receptors, targets, and pathways were proposed to provide insights into depression-related research and clinical treatment.
