植物乳杆菌细胞外囊泡的提取鉴定及细胞学功能验证

Extraction, identification and cytological function verification of extracellular vesicles from Lactobacillus plantarum

摘要: 为了探究植物乳杆菌来源的细胞外囊泡(LPEV)在细胞学方面的功能,采用超速离心法提取植物乳杆菌菌悬液中的细胞外囊泡,透射电镜观察囊泡形态特征;纳米颗粒跟踪分析(NTA)检测颗粒浓度;细胞增殖、细胞划痕、Transwell实验、荧光定量PCR、western blot免疫印迹分别验证该囊泡的细胞学功能。结果表明:自植物乳杆菌菌悬液分离出的细胞外囊泡,电镜下确实为囊泡状;浓度约为2.3×1010 颗粒数/mL;细胞增殖、细胞划痕、Transwell实验结果显示该囊泡可以促进细胞增殖,增强细胞活性;荧光定量PCR、western blot免疫印迹结果显示该囊泡对与皮肤再生有关的基因和蛋白的表达具有上调效果。综合实验结果证实该囊泡在再生医学方面具有一定的潜力,可以为后续探究相关具体调控机制奠定实验基础。

Abstract: To explore the cytological function of extracellular vesicles fromLactobacillus plantarum, extracellular vesicles fromL. plantarum(LPEV) suspension were extracted by differential centrifugation, the vesicles morphologic features were observed with a transmission electron microscope; the concentration was detected by nanoparticle tracking analysis (NTA); and the vesicles cytological function was verified by cell proliferation, cell scratch, Transwell, qRT-PCR and western blot. The results showed that the extracellular vesicles with the concentration about 2.3×1010 grain number/mL isolated fromL. plantarumwere indeed vesicle-like under electron microscope; the results of cell proliferation, cell scratch and Transwell showed that the vesicles could promote cell proliferation and enhance cell activity; qRT-PCR and western blot results showed that the vesicles had up-regulation effects on genes and proteins related to skin regeneration. The comprehensive experimental results confirmed that the vesicles had certain potential in regenerative medicine, which laid experimental foundation for further investigation of specific regulatory mechanisms.
