橡胶炭疽菌HD-1的鉴定及拮抗药剂筛选

Identification of Colletotrichum fructicola HD-1 and screening of antagonistic agents

摘要: 对实验室分离保存的一株橡胶炭疽菌(Rubber tree anthracnose)进行鉴定并对其拮抗药剂进行筛选,为橡胶树炭疽病的防治提供理论基础。通过分子生物学与形态学相结合的手段对该菌株进行鉴定,对其生物学特性进行研究,并且比较生防菌剂与化学药剂对该菌株的抑菌效果。结果表明:通过分子生物学与形态学将该菌株鉴定为果生刺盘孢菌(Colletotrichum fructicola),命名为HD-1。其最适生长的培养基、碳源、氮源分别为马铃薯葡萄糖琼脂培养基(PDA)、山梨醇和牛肉浸膏,最适pH值为8.0。通过测试和比较发现,咪鲜胺、苯醚甲环唑和贝莱斯芽孢杆菌(Bacillus velezensis)HN-2正丁醇提取物对菌株HD-1抑制效果明显,并且通过显微镜观察发现只有HN-2正丁醇提取物处理后的菌丝发生了膨大现象。实验结果将为对田间橡胶树炭疽病的防控提供理论依据和技术支撑。

Abstract: In this study, a strain of rubber anthracnose isolated and preserved in our laboratory was identified for antagonistic agents to provide a theoretical basis for the control of rubber anthracnose. The strain was identified by a combination of molecular biology and morphology, its biological characteristics were studied, and the inhibition effect of biological and chemical agents on the strain was compared. The results showed that this strain was identified as Colletotrichum fructicola and was named HD-1. The optimum medium, carbon, and nitrogen sources for growth were potato dextrose agar medium (PDA), sorbitol, and beef extract, with pH of 8.0. Through testing and comparison, it was found that Prochloraz, Difenoconazole, and Bacillus velezensis HN-2 n-butanol extracts had an obvious inhibitory effect on strain HD-1. Only the mycelium treated with HN-2 n-butanol extract was found to be expanded by microscopic observation. This experiment result would provide the theoretical basis and technical support for the prevention and control of rubber tree anthracnose in the field.
