Isolation and polygenic identification of rhizobia strains

Abstract: Two rhizobia strains, named T786 and T808, were isolated from the highland barley cultivation soil in Qamdo, Tibet, using R and M5 media at 4℃, respectively. After morphological, physiological, biochemical identification and 16S rRNA sequence analysis, strains T786 and T808 were preliminarily identified to genus Neorhizobium and Pararhizobium. The multiple sequence alignment results of 6 pairs housekeeping genes (atpD, recA, rpoB, thrC, glnII and glnA) in T786 and T808 showed that the similarity of these housekeeping genes and ANI values in strains T786 and T808 were lower than in the threshold of new species of rhizobia (95%-96%). Strains T786 and T808 were potential new species of rhizobia, the discovery of strains T786 and T808 was significance for enriching the germplasm resources of rhizobia in Qinghai-Tibet Plateau.
