摘要: 综述Ras相关域家族成员4 (Ras association domain family member 4, RASSF4)的分子结构、生物功能、基因启动子甲基化与肿瘤关系等研究进展,分析RASSF4具有调控细胞增殖、分化、凋亡等多个生物功能,其表达失调与多种肿瘤的发生发展的相关性。阐述启动子甲基化是RASSF4基因表达失调的重要机制之一,但RASSF4基因启动子甲基化与肿瘤发生发展的关联及意义尚不明晰,讨论以逆转甲基化的方式治疗肿瘤的应用前景。通过阐明RASSF4生物学概况和在多种肿瘤中所扮演的角色,为人类开发以RASSF4作为特定肿瘤的检测和治疗靶点提供理论依据。

Abstract: Ras association domain family member 4 (RASSF4) involves in multiple cell physiological activities such as cell proliferation, differentiation and apoptosis. The disordered expression of RASSF4 had shown significant correlation with tumors. To address this issue, the molecular structure, biological function, the relationship between promoter methylation and tumors were reviewed. One of the molecular mechanisms regulatingRASSF4gene expression is the DNA methylation of its gene promoter. However, the association and significance of the DNA methylation ofRASSF4gene promoter with the development of tumors are not clear. The application prospect of demethylation treatment of tumors was discussed. This review aimed to clarify the biological profile and role of RASSF4 in various tumors, thus providing a theoretical basis for the development of RASSF4 as a detection and treatment target for specific tumors.
