Test of expression pattern of tap gene in Drosophila melanoganster

Abstract: The tap gene of Drosophila melanogaster is characterized as ngn homologous gene because of its high base sequence similarity with mammal ngn gene. In the past two decades, the function and regulation of other genes except tap have been fully studied in the known ngn homologous group, while the corresponding research on tap gene has almost stagnated. Until used homologous recombination technique, the coding sequence of tap gene was accurately replaced with the coding sequence of Gal4 gene, and the tapGal4 strain was bred and cross with UAS-GFP strain. By fluorescence staining of GFP in Drosophila melanogaster, the expression pattern of tap gene in the development of the mushroom body was clearly observed. Furthermore, through quantitative analysis, it was determined that tap gene did not have proneural function in the development of mushroom body, but played an important role in the axon growth and right targeting. This paper mainly showed the expression pattern of tap gene in the development of brain and compound eye, which was helpful to knockout, knockdown or overexpress Tap protein in these organs, and then explore the function and regulation of tap gene in the development of these organs according to the resulting phenotypic changes.
