Abstract: To obtain Zizyphus jujube mill, Jinnan 1, tissue culture seedling contaminated with bacteria, methods were studied by using the ethanol, mercuric chloride and ultrasonic as primarily test factors together with the random block design of experiment. The results showed that optimal combined disinfection method for saving pollution tissue culture seedling was as follow samples soaked in 70% ethanol for 30 s, then in 0.1% mercuric chloride for 4 min and treated with ultrasonic(28 HZ,25℃) for15 min. Compared with the conventional ethanol-mercury chloride chemical disinfection method, pollution rate of inoculation explants used with this method was reduced 68.41%, the survival rate increased by 75.72%, the proliferation degree increased by 4.41 time. This a new method of physical and chemical disinfection showed good feasibility.
