Identification and analysis of O-mannosylated proteins from Bacillus Calmette-Guérin culture filter

Abstract: As the only licensed vaccine to protect against tuberculosis, Bacillus Calmette-Guérin(BCG)can not provide complete protection,therefore, it is essential to fully understand the antigens of BCG. The mannose containing glycoconjugates, which are located on cell wall, play a role in mycobacterial virulence and interaction with host cell. O-mannosylated proteins were reported in Mycobacterium tuberculosis and Mycobacterium smegmatis, but seldom in BCG. In this article, the O-mannosylated proteins from Bacillus Calmette-Guérin culture filter were identified and analyzed by ConA-agarose affinity chromatography combined with MALDI mass spectrometer. There were 7 kinds of O-mannosylated proteins identified and the mannosylated positions of these 7 proteins were further predicted. The identification and analysis of O-mannosylated proteins will shed light on immune mechanism of BCG.
