Genetic diversity of Larimichthys crocea based on mitochondrial DNA control region sequences in three cultured populations

Abstract: Genetic diversity of mitochondrial DNA control regions of three cultured Larimichthys crocea populations, common Min-yue(PM), Dai-qu(DQ) and "Min-you1"(MYOU), were analyzed by DNA sequencing technology. The results showed that the D-loop sequence length of the 98 individuals ranged from 785 to 799bp. The total number of haplotypes was 48, in which 18 were in both common Min-yue and "Min-you1" , and 17 in Dai-qu. The effective numbers of polymorphic loci were 167, and insertion/deletion loci were 39 in three populations. The average of convert/transversion is 6.33. Although the average number of haploid type diversity (Hd) was 0.8026, the nucleotide diversity (π) remains low. The π volumes of three populations, PM, DQ and MYOU, were 0.01468, 0.00266 and 0.01208, respectively, and the average number of π was 0.00981. The results indicated that the genetic diversity of DQ population is far lower than PM .It was suggested that the effect of selection on genetic diversity should be taken care.
