Abstract: The analysis of chromosome karyotype is one of the basic methods for the study of cytogenetics and genetic breeding. As a carrier of genes, chromosome has species-specific, which are of great significance for studying of the evolution classification of species and the relationship among the structures morphology and functions of chromosome. The tender leaves of the 6th generation of test tube seedlings obtained from anther culture of Zizyphus jujube mill Jinnan No.1 were selected as materials, the enzyme solution to wall low permeability-modified phenol fuchsin staining method was used to make specimen slice and auxiliary microscopic observation analysis was used to study its karyotype. The results showed that the number of chromosomes in this test tube seedlings were 12, including 1 chromosome group and the small chromosome. The formula of chromosome group is n=1X=12=9m+3sm, belonging to type 2B. In theory, this study would provide genetic basis for the rapid propagation and breeding of anther culture regeneration plant from this Chinese jujube. At the same time, the improved method for the analysis of jujube chromosome group was provided.
