Analysis of relationship between DNA fingerprinting by RAPD and species of spring plankton in Hengshan reservoir

Abstract:  The RAPD fingerprinting based on the DNA polymorphism of the plankton communities at 12 sampling points from 4 stations in Hengshan Reservoir, Zhejiang, was analyzed and the species were identified in April 2013. The relationship between topological structure of DNA fingerprinting and species was discussed by the cluster analysis. The results showed that 87 bands with the length ranging from 120 to 1700 bp were gained from 12 screened random primers, and the polymorphic rate was 87.4%. On average, 56.2 bands were observed from each station. A maximum number of 63 bands were observed from the lower level of station 1 and a minimum of 50 bands were observed from the middle level of station 4. 64 kinds of planktons were identified from 4 stations with the maximum number of 27 in the upper level of station 4 and the minimum of 16 in the upper level of station 3. 17-25 kinds of planktons were observed from other stations. Achnanthes sp., Ankistrodesmus convolutus, Chlorella vulgaris, Phormidium tenue and Tintinnidium sp. were all detected from 12 sampling points. The plankton communities from 12 sampling points can be divided into two types by systematic cluster analysis based on the species and RAPD. This study reveals a close relationship between the DNA fingerprinting structure of the plankton community and species in Hengshan Reservoir.
