Abstract: In order to diagnose and monitor FMD(foot-and-mouth disease) efficiently and accurately, this study established a high throughput liquid-phase blocking CLIA antibody detection method of the O-type FMD. Through using one single dilution instead of the gradient dilution, the detection flux was raised about 5 times. The test results were assessed by percentage inhibition, and the optimal cut-off value was 62.5% determined by the receiver operating characteristic curve (ROC). Repeatability test showed the intro batch CV was under 10% and the inter batch CV was under 15%. By comparing the results of 326 field serum samples with those of LPB-ELISA, the sensitivity, sepecificity and accuraly were 94.6%, 95.6% and 95.1%, respectively. The results showed that the kit reduce the time and economic cost on the basis of the good sensitivity, specificity and reliability of the results. It also provides a new method for the quarantine of foot and mouth disease in China.
