Enhanced endo-β-1,3-glucanase produciton through morphology control of Trichoderma harzianum

Abstract: There is a close relationship between morphology of Trichoderma harzianum and endo-β-1,3-glucanase production. The culture conditions for good morphology of Trichoderma harzianumin was optimized by single factor experiment, and the result showed as follow: 100 mL medium in 500 mL Erlenmeyer flask, 5% (V/V) of inoculum volume. The initial pH of the medium was 6.5 and shaking speed was 180 r/min, 0.3%(m/V) Tween-80 or 0.3%(m/V) CMC-Na was added into the medium as dispersing agents. The mycelia pellet diameter was about (0.42±0.09) mm and the activity of endo-β-1,3-glucan was (111.9±7.5) U/mL. In the further orthogonal optimization, the activity of endo-β-1,3-glucan reached (135.2±8.3) U/mL, and the influence order of culture conditions to mycelia pellet diameter was inoculum volume >CMC-Na> Tween-80.
