镉诱导不同自噬效应与钙离子调控自噬的作用分析

The relationship between cadmium-induced different autophagy effect and calcium-regulated autophagy

摘要: 环境中重金属镉污染威胁人类健康。镉在人体中长期积累,对机体的多种器官和系统造成损伤,如肝肾组织、免疫系统、心血管系统等。对镉毒性机制的研究主要集中在氧化损伤、DNA损伤、细胞凋亡等方面。近年来,越来越多的研究结果认为镉对细胞自噬的不同影响与细胞存活之间存在复杂的关联。镉诱导的细胞自噬在不同细胞中表现为抑制或促进细胞凋亡;镉也可通过阻碍自噬体与溶酶体的融合,抑制自噬流、从而诱导细胞凋亡。镉通过干扰细胞中溶酶体及钙离子的代谢影响自噬过程。综述了镉在不同细胞中引发不同自噬效应,探究其可能的分子机制。

Abstract: Pollution of the environment by the heavy metal cadmium poses a great threat to human health. The long-term accumulation of Cd can cause damage to various organs and systems of the body, such as liver and kidney tissues, immune system and cardiovascular system, etc. The research on toxicity mechanism of Cd mainly focuses on oxidative damage, DNA damage and cell apoptosis. Studies in recent years have suggested that the different effects of Cd on autophagy have a complex relationship with cell survival. Cd is known to induce autophagy which inhibit or facilitate cell apoptosis in different cells. It can also inhibit autophagy flux and induce apoptosis by blocking the fusion of autophagosome and lysosome. Cd affects autophagy by interfering with the lysosomes and metabolism of calcium ions in cells. This paper reviews the different autophagy effects induced by Cd in different cells, and explores its possible molecular mechanisms.
