摘要: Clusterin(CLU),是一种存在多种细胞组分中具有多种存在形式、功能复杂多样的蛋白,最早是在1983年由Blaschuk报道存在于山羊精液中促进细胞间黏附的糖蛋白。随后的研究发现,该蛋白参与细胞内外重要的生命活动,并且与包括神经退行性疾病、免疫疾病、衰老、肿瘤等在内的许多疾病相关。CLU的功能多样,甚至在相互对立的方面均发挥重要的功能,使研究者们感到十分迷惑。目前认为,clusterin可以编码不同的蛋白产物,具有不同的细胞定位,发挥不同的生物学功能,主要是分泌型(sCLU)及核型(nCLU)。就CLU不同蛋白产物的结构、定位、产生机制、具有的功能以及参与的信号通路做一综述。

Abstract: Clusterin (CLU) is a multifunctional protein existing in distinct subcellular components. It was first reported in 1983 as an adhesion glycoprotein in goat semen by Blaschuk, then was found involved in numerous physiological processes and associated with many pathological states (neurodegenerative diseases, immune diseases, aging, cancer, etc.). CLU plays different roles, and even in opposing aspect, which confused researchers. To summarize results of different research groups, we will have a more clear understanding of this protein. Currently, it is considered that clusterin may encode different protein products (primarily secreted CLU (sCLU) and karyotype CLU (nCLU)), with different cellular localization and play different biological functions. In this paper, the structure, function, generating mechanism and the signal network of this protein will be reviewed.
