腐乳来源乳酸菌的分离及其对斑马鱼生长发育的影响

Influence of lactic acid bacterium derived from fermented beancurd on growth and development of zebrafish

摘要: 益生菌对动物健康具有重要作用,但对动物幼体的摄食和运动能力的影响尚不明确。探讨益生菌对动物体生长发育的影响,对于幼体营养调控具有重要的意义。传统发酵食品是益生菌的来源之一,研究利用乳酸菌培养基(MRS),从腐乳中分离筛选得到一株耐胆盐的乳酸菌(FR27),经测序该菌属于植物乳杆菌(Lactobacillus plantarum)。为了探究FR27对斑马鱼生长发育的影响,收集斑马鱼受精卵,并将其分为3个处理组,对照组(不加菌)、FR27低浓度添加组(水体加菌终浓度104cfu/mL)和FR27高浓度添加组(水体加菌终浓度105cfu/mL),实验持续3周。在实验周期内测量统计斑马鱼卵孵化率、幼鱼生长发育以及行为学相关指标,结果表明FR27的添加能够提高斑马鱼卵的孵化率、增加幼鱼体长和脂肪沉积,并且高浓度组更显著(P<0.05)。通过摄食实验和行为仪检测,可以发现FR27添加组幼鱼的摄食和运动能力显著提高,高浓度组与低浓度组相比,摄食和运动能力提高更显著。综上,推测FR27通过增强斑马鱼幼鱼的运动能力和摄食能力,促进幼鱼生长发育和脂肪沉积。

Abstract: Probiotics play an important role in animal health, but the effects on the feed intake and moving capacity of animal larvae are not clear. Exploring the effects of probiotics on the growth and development of animals is of great significance for the regulation of larval nutrition. Traditional fermented foods are one of the sources of probiotics. In this study, a lactic acid bacteria (FR27) resistant to bile salts was isolated from fermented bean curd by using lactic acid bacteria culture medium (MRS). The strain was sequenced and the results showed this strain was affiliated to Lactobacillus plantarum. To explore the effect of FR27 on the growth and development of zebrafish, the fertilized eggs of zebrafish were collected and divided into three treatment groups, the control group (without adding bacteria), the low-concentration group (adding 104 cfu/ mL FR27) and the high-concentration group (adding 105 cfu/mL FR27), and the treatment lasted for three weeks. The hatching rate of zebrafish eggs, the growth condition and development of juvenile fish were measured. The results showed that the addition of FR27 could increase the hatching rate of zebrafish eggs, the body length and fat deposition of juvenile fish, which were more significant (P<0.05) in the high-concentration group as compared with the other groups. The feed intake and movement of juvenile fish were increased in FR27-added group. In summary, we speculated that FR27 promotes juvenile fish growth and fat deposition by enhancing the feed intake and moving ability of zebrafish juveniles.
