光唇鱼早期性腺组织学观察及激素诱导性逆转评价

Histological observation of early gonadal development and evaluation of hormone induced sex reversal in Acrossocheilus fasciatus

摘要: 为比较光唇鱼雌雄差异性,对199尾16月龄光唇鱼的生长性状(体长和体质量)进行雌雄差异性统计分析,采用组织学切片技术对光唇鱼早期性腺及外源性激素诱导的性腺进行研究,并对性别分化相关基因表达进行定量分析。结果显示,光唇鱼在生长性状方面存在明显的性别二态性,16月龄雌鱼平均体长较雄鱼增加约9.6%,平均体质量高出雄鱼33.7%。组织学切片结果显示,19 dph时初次观察到光唇鱼的原始性腺,卵巢在48 dph已经分化,精巢分化晚于55 dph。对7 dph的仔鱼利用2种浓度(50 mg/kg和100 mg/kg)雌雄激素进行60 d的性逆转诱导处理,结果表明,雄激素甲基睾酮(MT)-100组(100 mg/kg)的雄鱼占比为82%,诱导效率优于MT-50组(50 mg/kg),雌激素雌二醇(E2) 2种浓度处理组的雌鱼占比均达100%。MT组的精巢发育相关基因dmrt1和amh的表达均上调,而卵巢发育相关基因cyp19a1a和foxl2的表达下调;E2组的cyp19a1a和foxl2的表达上调,而dmrt1和amh的表达则下调。实验结果鉴定出光唇鱼卵巢分化的时间,同时建立光唇鱼性逆转方案,为光唇鱼性别分化机制以及单性育种研究提供参考资料。

Abstract: To compare the difference between the male and female individuals of Acrossocheilus fasciatus, the variations of growth traits (body length and body weight) were investigated among a total of 199 16-month-old individuals, and the early gonadal development and the exogenous hormone-induced gonad in A.fasciatus were studied using histological sectioning techniques and the sex differentiation-related genes were quantified. The results showed that there was obvious sexual dimorphism in growth traits, and the average body length of 16-months-old females increased by about 9.6% compared with that of the males, and the average body mass of the females was 33.7% higher than that of the males. The results of histological sections showed that primordial gonad was initially observed at 19 dph and it differentiated into ovary at 48 dph. However, the differentiated testis appeared later than 55 dph. The sex reversal induced by sex hormones with two different concentration (50 mg/kg and 100 mg/kg) lasting for 60 days was performed on 7 dph larvae of A.fasciatus. The results showed that the percentage of the males in the methyltestasterone (MT)-100 group (100 mg/kg) was 82%, which was higher than that in the MT-50 group (50 mg/kg), and the estradiol (E2) treatment group had 100% of the females at both concentrations. In addition, the expression level of the male differentiation-related genes dmrt1 and amh was up-regulated and the expression level of the female differentiation-related genes cyp19a1a and foxl2 was down-regulated in the MT group. While the expression level of cyp19a1a and foxl2 was up-regulated and the expression of dmrt1 and amh was down-regulated in the E2 group. In this study, the time point of ovarian differentiation was identified and a sex reversal program was also established in A.fasciatus, which would provide information and reference data for further research on the mechanism of sex differentiation as well as unisexual breeding in A.fasciatus.
