基于超高压技术的牛骨蛋白绿色提取工艺初探

Preliminary study on green extraction technology of bovine bone protein based on ultra high pressure technology

摘要: 为研究绿色牛骨蛋白提取工艺,利用超高压技术在温和环境中破坏骨结构,加速牛骨蛋白释放。以脱钙牛骨粉为实验对象,通过单因素和响应面优化实验,研究保压时间、作用压力和环境pH在牛骨蛋白超高压提取工艺中对蛋白提取率的影响,得出超高压提取牛骨蛋白的最佳条件。响应面实验回归模型显著,误差较小,模型适用于本实验理论解释。结果表明,在保压时间54 min、作用压力189 MPa和环境pH 6.7时蛋白提取率最高,可达35.62%,与理论模型预测值吻合度达98.8%。将提取次数增加到3次,提取率可达54.2%,结合酶提取率可达59.3%。建立超高压提取牛骨蛋白工艺,实现牛骨蛋白的绿色提取。

Abstract: To research environment-friendly extraction method, ultra-high pressure method was used to destroy bone structure in nature environment and to accelerate the release of bovine protein. The effects of time, pressure and pH on the extraction rate of protein from bovine bone meal were studied by single factor and response surface optimization experiment. The optimal conditions for extraction of bovine bone protein under ultra-high pressure were obtained. The regression model of response surface experiment was significant and the error was small. The model was suitable for theoretical interpretation of this experiment. The results showed that the protein extraction rate reached 35.62% when time was 54 min, pressure was 189 MPa and pH was 6.7. When the number of extraction was increased to three times, the extraction rate reached 54.2%. And it could reach 59.3% when hydrolyzed with enzyme. It was established an ultra high pressure the environment-friendly bovine bone protein extraction method.
