中缅树鼩Cyt b基因遗传多样性的研究

Genetic diversity of Cytochrome b gene in Tupaia belangeri

摘要: 中缅树鼩是东洋界特有的小型哺乳动物,基于11个地理种群的中缅树鼩研究其Cyt b基因的遗传多样性。基于MEGA软件共获取Cyt b基因有效序列1073 bp,124个样本中A+T(55.8%)的比例高于C+G(44.2%),其中发现变异位点数为262个,共定义了66种单倍型。基于Kimura两参数邻接法构建的系统发育树显示,11个种群的中缅树鼩聚为4大支:大新种群一支、海南种群一支、片马和腾冲种群一支,其余种群(昆明、兴义、大理、勐腊、河口、西昌、乐业)聚为一支。AMOVA分析表明种群间的分子变异占总变异的75.12%,种群内的分子变异占总变异的24.88%,表明变异主要发生在种群间。各种群间的基因流均小于1,推测中缅树鼩可能发生了一定的种群分化。中缅树鼩中性检测的Tajima′s D和错配分析检验结果均不显著,表明中缅树鼩在历史进程中没有经历过种群扩张。

Abstract: Tupaia belangeri is a unique small mammal of the Oriental realm. In the present study, the genetic diversity base on Cyt b gene (1073 bp)of 11 geographical populations in Tupaia belangeri was analyzed. The ratio of A+T (55.8%)was higher than that of C+G (44.2%)in the 124 samples, and 262 variable sites and 66 haplotypes found. Phylogenetic tree was built based on Kimura two-parameter Neighbor-joining method, and 11 populatians were divided into 4 branches: Daxin population, Hainan population, Pianma and Tengchong population, and the other populations(Kunming, Xingyi, Dali, Mengla, Hekou, Xichang, Leye). AMOVA analysis showed that variations inter-populations and intra-populations were 75.12% and 24.88%, respectively. The variations mainly occurred among populations and the gene flow between populations was less than 1. All of the result indicated that Tupaia belangeri might have a certain population differentiation. The Tajima′s D and Mismatch distribution test results of neutral detection in Tupaia belangeri were negative, suggesting that Tupaia belangeri has not experienced population expansion in history.
