乌贼墨汁黑色素对小鼠体内重金属的脱除作用

Removal of heavy metals in mice by melanin from sepia

摘要: 研究了乌贼墨汁黑色素对小鼠体内重金属的脱除作用。将小鼠随机分为4组,正常对照组、黑色素组、重金属组、重金属和黑色素组,分别灌胃生理盐水、黑色素悬浊液、重金属溶液、重金属和黑色素混合液。连续灌胃7 d后,计算脾脏指数,测定小鼠血液、股骨和粪便中重金属铅(Pb)、镉(Cd)、铬(Cr)、钡(Ba)含量,以及肝脏中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活力、丙二醛(MDA)含量。结果显示,灌胃重金属和黑色素后,与重金属组相比,Pb、Cr、Ba在血液、股骨中含量显著下降,在粪便中含量显著上升;Cd在血液、股骨中含量也降低,在粪便中含量显著上升。灌胃黑色素后,与正常组相比,脾脏指数变化不显著;SOD、GSH-Px活力下降,MDA含量上升。提示乌贼墨汁黑色素对Pb、Cd、Cr、Ba有减少吸收,促进排出的作用,具体机制有待进一步研究。

Abstract: In this study, we used mouse model to analyze the removal of heavy metals by melanin extracted from sepia. The mice were randomly divided into 4 groups: as the control group, melanin group, heavy metals group, heavy metals and melanin group, which were treated by gavage with saline, melanin, heavy metals, heavy metals and melanin, respectively. After a trial period of 7 days, we calculated the data of experimental animals, such as spleen index, heavy metal(Pb,Cd,Cr and Ba) contents in different organs, activities of enzymes (SOD,GSH-Px), and the content of MDA. The results showed that Pb, Cr and Ba contents decreased significantly in blood and femur, and increased significantly in feces after heavy metals and melanin were administered compared with the heavy metals group. Besides, in contrast with heavy metals group, Cd content in of the heavy metals and melanin group dropped in blood and femur, and significantly rised in feces as well. As for melanin group, it had no significant change in spleen index by comparison with the control group. The activities of SOD, GSH-Px in melanin group were lower than the control group while the content of MDA was higher. Thus, sepia melanin appeared to help reduce Pb, Cd, Cr and Ba absorption and promote excretion.
