预过滤孔径大小显著影响浮游细菌群落分析结果

Effect of pre-filtering pore-size significantly on the results of the analysis of planktonic bacterial communities

摘要: 为探究预过滤孔径对浮游细菌群落结构的影响,通过高通量测序技术,比较了100 μm(PF100)、10 μm(PF10)和2 μm(PF2)3种不同孔径的预过滤处理后对虾养殖水体浮游细菌群落结构。结果显示,PF100、PF10与PF2处理下的浮游细菌群落结构和多样性存在显著差异(P<0.05),且PF100的细菌群落多样性最高,但PF10和PF2的细菌群落结构和多样性差异不显著(P>0.05)。PF100的绿弯菌门(Chloroflexi)、浮霉菌门(Planctomycetes)、疣微菌门(Verrucomicrobia)和蓝细菌门(Cyanobacteria)的相对丰度显著高于PF10和PF2(P<0.05),相反,α-变形菌纲(Alphaproteobacteria)、GN02和OD1在PF10和PF2中的相对丰度较高。相似性百分比分析(SIMPER)结果表明,腐螺旋菌科(Saprospiraceae)和黄杆菌科(Flavobacteriaceae)等12个细菌科是造成3种预过滤处理细菌群落差异的主要类群。综上所述,预过滤孔径大小会导致浮游细菌群落结构的差异,该结果对浮游细菌过滤方法的选择具有一定指导意义。

Abstract: To study the effect of different pre-filtering pore-size on planktonic bacterial community compositions (BCCs), BCCs collected by three filtering methods (10 μm pre-filtering, 2 μm pre-filtering, and without pre-filtering) were compared through high-throughput sequencing. The result showed that BCCs were similar between 10 μm pre-filtering (PF10) and 2 μm pre-filtering (PF2), while they were significantly different with those without filtering (PF100). The alpha diversity of PF100 samples was the highest among all the treatments. Compared with PF10 and PF2, the relative abundances of Chloroflexi, Planctomycetes, Verrucomicrobia and Cyanobacteria were significantly higher in PF100 samples (P < 0.05), while those of Alphaproteobacteria, GN02 and OD1 were lower. Similarity percentage (SIMPER) analysis indicated that 12 families cumulatively contributed to 63.45% dissimilarity of the BCCs. In conclusion, the different pre-filtering pore-size could induce the bias of planktonic bacterial community structure. The results have an important guiding significance in selecting the planktonic bacteria filtration method.
