香鱼成熟卵母细胞过熟进程中卵膜超微结构变化

Changes of egg envelope of sweet fish Plecoglossus altivelis during post-ovulatory ageing

摘要:

香鱼成熟卵母细胞能较长时间维持受精能力,但随时间延迟受精能力逐渐降低直至消失。为探索其机制,采用透射电镜技术观察了香鱼卵母细胞过熟进程中卵膜超微结构的变化。结果显示:处于滤泡内时,成熟卵母细胞表面的放射膜(Zona radiate, ZR)分为3层,从外到内依次是放射外膜1(ZR1)、放射膜2(ZR2)、放射膜3(ZR3),其中ZR1最厚;放射膜外有一层胶膜(Jelly coat, JC)。排出后的成熟卵母细胞放射膜ZR1消失,仅ZR2和ZR3两层,但放射膜外面的胶膜层依然存在,此时ZR2很薄,ZR3层次明暗分明,包含明暗相间的各5条带并且完全不透明,在与卵膜垂直的方向上形成亮区(Light region, LR)和暗区(Dark region, DR)。随着排卵后的时限延长,至48 h后,卵的胶膜层消失,放射膜压缩,内部出现孔道(Pore canals, PC);60 h后胶膜层孔道数量增多,孔径变大;至96 h时孔道变长,贯穿ZR3的1/2;至104 h时ZR2基本解体,孔道继续伸长并贯穿ZR3,至此成熟卵母细胞彻底凋亡。

Abstract:

The mature oocytes of sweet fish Plecoglossus altivelis can maintain fertilization ability for a long time. In this study, we observed the ultrastructure′ changes of P. altivelis oocytes during post-ovulatory process using transmission electron microscope(TEM). In follicular phase, the outer layer of oocyte memebrane is zona radiate(ZR), which includes three-layers(ZR1, ZR2 and ZR3 from outside to inside, and the thickest layer is ZR1). Outside of ZR is jelly coat(JC). After ovulation, ZR1 disappears and ZR remains two layers (ZR2 and ZR3), and JC still exisst. In this stage, ZR2 is very thin, and ZR3 includes 5 light-bands alternate with 5 dark-bands. ZR3 is completely opaque, and in the vertical of oolemma showed alternating "light region" and "dark region". At 48 h after ovulation, JC disappears, ZR is thinner and pore canals(PC) appears in ZR3. At 60 h, the number of PC increases and diameter enlarges. To 96 h, the length of PC increases to 1/2 of the ZR3. 104 h later, with the disintegration of ZR2, PC continues to elongate and pass through ZR3, and then the mature oocyte completely apoptosis.
