草铵膦对四膜虫毒性作用的研究

Research on the toxicity of glufosinate ammonium to Tetrahymena thermophila

摘要: 从细胞增殖和DNA损伤角度研究不同浓度的草铵膦对嗜热四膜虫(Tetrahymena thermophila)的毒性作用。采用12 h间隔细胞计数绘制96 h生长曲线,单细胞凝胶电泳技术检测受试四膜虫的尾长(TL)、尾矩(TM)及Olive尾矩(OTM)。生长曲线绘制实验表明:草铵膦处理组与空白组种群细胞数目差异显著(P<0.01)。单细胞凝胶电泳实验表明:染毒各组的细胞TL、TM、OTM与对照组相比,均有显著增大(P<0.01),DNA损伤级别随着染毒浓度增大而升高(P<0.01)。实验浓度的草铵膦对细胞DNA均造成了不同程度的损伤,反映了其对四膜虫具有急性遗传毒性

Abstract: The toxicity effects of glufosinate ammonium to Tetrahymena thermophila were investigated from the viewpoints of cell proliferation and DNA damage. The growth curves of 96 hours were plotted by counting the cells every 12 hours; the tail length, tail moment and Olive tail moment of Tetrahymena thermophila were measured by Single Cell Gel Electrophoresis (SCGE). The results of growth curve test showed that there was a significant difference between drug groups and control group in cell density (P<0.01). According to SCGE data, the tail length, tail moment and Olive tail moment of the tested series were all significantly larger and higher than those in the control group (P<0.01), the DNA damage level was higher when toxic compound become heavier (P<0.01). The experimental concentration of glufosinate ammonium caused damages of cell DNA in varied extents, which reflects the acute and genetic toxicity of it to Tetrahymena thermophila .
