通识选修课开设微丝荧光观察实验的探究

Research on establishment of experiment for observing of microfilament by fluorescence labeling in general elective course

摘要: 课程组在通识选修课基础生命科学导论实验中开设了以洋葱鳞茎为试验材料的微丝荧光观察实验。实验使用鬼笔环肽-罗丹明标记法对洋葱内表皮细胞中的微丝进行标记,在荧光显微镜下观察到洋葱内表皮细胞中的微丝发出红色荧光,微丝相互交织,充满整个细胞,部分位置有分叉,在细胞核周围分布较为密集。针对通识选修课的特点,使用洋葱代替动物细胞作为试验材料,降低了实验操作的难度,增强了非生物类专业学生对生命科学前沿知识和实验技术的了解,培养学生对生命科学的兴趣,同时丰富了课程的实验内容,提升了课程的教学质量,取得了良好的教学效果。

Abstract: The course group takes onion bulbs as the experimental materials to establish the experiment of observing of microfilaments by fluorescence labeling in the general elective course of the introduction to basic life science experiment. In this experiment, the microfilaments in the inner epidermis cells of onion are labeled by the phalloidin-rhodamine. Under the fluorescence microscope, the students can observe that the microfilaments in the onion inner epidermal cells emit red fluorescence. The microfilaments fill the whole cell and are interweaved with each other. Around the nucleus, the microfilaments are denser. According to the characteristics of general elective course, onion is used instead of animal cells as experimental material, which reduces the difficulty of experimental operation. The experiment helps the non-biological majors to enhance their understanding of the frontier knowledge and experimental technology of life science, and cultivate their interest in life science by interpreting the basic knowledge of cytoskeleton, microfilaments and fluorescent labeling technology and training the experimental method of labeling microfilaments withphalloidin-rhodamine. At the same time, it enriches the experimental contents, improves the teaching quality of the course and obtains good effects.
