Expression and location of 5-hydroxytryptamine and dopamine in the cerebellum of the new-hatching Yangtze alligator Alligator sinensis

摘要: 运用Nissl和免疫组织化学染色方法研究了初孵扬子鳄小脑的组织学结构以及5-羟色胺(5-HT)和多巴胺(DA)在初孵扬子鳄小脑的表达和定位情况。Nissl染色结果显示,初孵扬子鳄的小脑有小脑室,小脑皮质表面不光滑、有皱褶,可见沟回分化。小脑髓质内可见小脑核,为外侧小脑核和内侧小脑核。免疫组织化学染色结果显示,小脑皮质可分为4层,由外向内依次是:外颗粒层、分子层、浦肯野细胞层和颗粒层。初孵扬子鳄小脑皮质的浦肯野细胞为多细胞层。冠状切面上,外颗粒层细胞呈5-HT免疫阳性和微弱的DA免疫阳性;浦肯野细胞呈5-HT免疫阳性和DA免疫阳性;分子层和颗粒层均显示5-HT神经纤维免疫阳性且阳性较强,显示DA神经纤维免疫阳性;髓质有较多DA免疫阳性纤维且分布密集;髓质内核团细胞体呈5-HT免疫阳性,但呈DA免疫阴性;第四脑脑室脑脊液物质也显示DA免疫阳性。矢状切面上,可见外颗粒层细胞5-HT免疫阳性明显,外颗粒层纤维DA免疫阳性明显;分子层和颗粒层神经纤维呈5-HT免疫阳性和DA免疫阳性;小脑室的室管膜细胞呈5-HT免疫阴性和DA免疫阴性;脉络丛细胞呈5-HT浅阳性和DA强阳性。5-羟色胺和多巴胺在小脑的广泛分布表明其在运动控制等方面发挥了重要作用。

Abstract: Nissl staining and immunohistochemical staining method were used to morphologically characterize the cerebellum of the new-hatching Yangtze alligator Alligator sinensis, and the distribution of 5-HT and DA in the cerebellum of the new-hatching Yangtze alligator was investigated for the first time. The results showed that there was a ventriculus cerebelli in the cerebellum of the new-hatching Yangtze alligator and the surface of the cerebellar cortex was not very smooth but with irregular folds. Two cerebellar nuclei termed as the nucleus cerebelli lateralis and the nucleus cerebelli medialis were found in the medulla of cerebellum. The results also showed the cerebellar cortex could be divided into four layers which included external granular layer (EGL), molecular layer (ML), Purkinje cell layer (PCL) and granular layer (GL). Purkinje cell layer could be characterized specially by multilayer. In the transverse sections, the 5-hydroxytryptamine-immunoreactive (5-HT-IR) and dopamine-immunoreactive (DA-IR) cells were detected in the EGL; the 5-HT-IR and DA-IR Purkinje cells were also found; there were 5-HT-IR fibers in the ML and GL, the DA-IR fibers could also be found in the ML and GL; a lot of 5-HT-IR and DA-IR fibers were detected in the medulla; the cell bodies of cerebellar nucleus were also detected 5-HT-IR; the substance of CSF was found DA-IR. In the sagittal sections, there were 5-HT-IR cells and DA-IR fibers in the EGL; there were also 5-HT-IR and DA-IR fibers in the ML and GL; the ependymal cells were not detected 5-HT-IR and DA-IR; the cells of the choroid plexus (CP) were found lightly 5-HT-IR and strongly DA-IR. The abundance of 5-HT and DA in cerebellum suggested that these highly conserved neurotransmitters would play important roles in motor control.
