低产桔霉素红曲霉复合诱变育种

Screening of low-yield citrinin monascus by compound mutation breeding

摘要: 以通过N+离子束诱变筛选出的高产洛伐他汀红曲霉菌株M50-2为出发菌株,进行紫外诱变,筛选低桔霉素产量菌株。将出发菌株置于30 W紫外灯下30 cm处分别照射30、60、90、120和150 s后,对所得诱变菌株进行液态发酵。采用高效液相色谱法(HPLC)中的荧光检测(λex=331 nm,λem=500 nm)和紫外检测(λ=238 nm)测定发酵产物中的桔霉素和洛伐他汀的含量。结果表明,通过紫外诱变,获得一株优良菌株。其液态发酵的产物中的桔霉素产量为2.15 mg/L,与出发菌株相比桔霉素含量减少了96.6%,而洛伐他汀产量为0.338 mg/mL,与出发菌株相比只减少了16.5%,对该菌株进行传代培养12代,结果表明该菌株具有较好的遗传稳定性。

Abstract:  In order to screen out low citrinin yield strain, as a starting strain, the high-yield lovastatin Monascus strain M50-2 was treated by ultraviolet, which was selected by N+ ion beam mutagenesis. The starting strain was irradiated at 30 cm under a 30 W UV lamp, and the dose was 30, 60, 90, 120 and 150 s. The contents of citrinin and lovastatin in the fermentation products were determined by high performance liquid chromatography (HPLC) with fluorescence detection (λex=331 nm, λem=500 nm) and ultraviolet detection (λ=238 nm) after the liquid fermentation of the screened mutagenized strain. The results showed that an excellent strain M120-1 was obtained by ultraviolet mutagenesis. The best strain M120-1 yielded 2.15 mg/L citrinin, which was 96.6% lower than the original strain. However, lovastatin production was 0.338 mg/mL, which was only decreased by 16.5%. To examine the genetic stability of M120-1, the strain was continuously sub-cultured to the 12th generation, showing that the strain had good genetic stability.
