白藜芦醇抑制肝癌Smmc-7721细胞增殖及机制研究

The inhibitory effect of resveratrol in hepatocellular carcinoma Smmc-7721 cells

摘要: 采用MTT法研究白藜芦醇对肝癌Smmc-7721细胞增殖的影响,并采用Western blotting法分析p-Erk1/2/Erk1/2、p-mTOR/mTOR和p-Akt/Akt蛋白水平。结果表明:白藜芦醇处理肝癌Smmc-7721细胞后,细胞活力受到明显抑制(P< 0.001)。80 μM白藜芦醇处理肝癌Smmc-7721细胞24 h时,细胞的p-Erk1/2/Erk1/2蛋白水平明显降低(P = 0.009)。且白藜芦醇处理肝癌Smmc-7721细胞72 h时,p-mTOR/mTOR蛋白水平明显提高,但p-Akt/Akt蛋白水平却无明显差异。白藜芦醇可通过Erk1/2信号途径抑制肝癌细胞增殖。同时,白藜芦醇可通过Akt非依赖的途径激活mTOR信号通路,提示白藜芦醇可调控肝癌Smmc-7721细胞的能量代谢。

Abstract: To investigate the inhibitory effect and mechanism of resveratrol in hepatocellular carcinoma cells, cell viabilities were detected by MTT assay and protein levels were detected by Western blotting. The results indicated that resveratrol significantly exhibited inhibitory effect in hepatocellular carcinoma Smmc-7721 cells. And the level of p-Erk1/2/Erk1/2 significantly decreased in cells treated with resveratrol. Furthermore, the level of p-mTOR/mTOR significantly increased in cells treated with resveratrol. But, there was no significant change in the level of p-Akt/Akt. The data suggest that resveratrol exhibits the inhibitory effect via Erk1/2 pathway in hepatocellular carcinoma cells. And resveratrol can regulate the energy metabolism via mTOR pathway in hepatocellular carcinoma cells.
