Inhibitory effects and preliminary molecular mechanisms of “natural plant antimicrobial solution”(PAMs)on liver cancer HepG2 cells

摘要: 肝癌是一类严重危害人类生命的重大疾病。前期研究发现,传统中药和民族药“天然植物抗菌液”(PAMs)对白血病、银屑病、炎症等多种疾病具有广泛的生物学活性与作用,但其对肝癌细胞,尤其是分子作用机制尚不清楚。研究确认,PAMs抑制HepG2细胞活力并呈现时间和浓度依赖性;PAMs上调caspase3、caspase8、caspase9、p53并同时下调Bcl-2的表达。进一步的HepG2细胞裸鼠移植瘤实验表明,仅仅体外涂搽PAMs即可通过下调FoxM1的表达而显著抑制肿瘤的生长并促进细胞凋亡。作为一种安全、有效、方便的候选药物,PAMs可望用于肝癌等多种疾病的预防和治疗。

Abstract: Liver cancer is a critical disease that seriously injures the human life. Previous studies revealed that the traditional and folk Chinese medicine "natural plant antibacterial solution" (PAMs) has wide biological activities and effects on many diseases such as leukemia, psoriasis and inflammation. However, its effect on hepatoma cells, especially the molecular mechanism is not well understood. In this study, we confirmed that PAMs could inhibit the viabilities of HepG2 cells dependent on the concentration and time. PAMs enhanced the expression of caspase3, caspase8, caspase9 and p53, while it repressed the expression of Bcl-2. Furthermore, the nude mice HepG2 cell xenograft assay showed that PAMs significantly inhibited the tumor growth and promoted the apoptosis of HepG2 cells by downregulating FoxM1 expression merely by perfrication. Being a safe, effective and convenient candidate drug, PAMs is hoped to be used in the prevention and therapy of various diseases including liver cancer.
