BDNF对中枢神经系统疾病作用的研究进展

摘要: 介绍脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)在中枢神经系统(central nervous system,CNS)功能及相关疾病中的重要作用,包括促进神经元生长和存活、增强突触可塑性、参与神经递质调节、促进髓鞘形成以及神经保护等功能,提示BDNF对改善和治疗CNS疾病有重要作用。通过总结与BDNF相关的动物实验和临床前研究,概述BDNF对脊髓损伤、创伤性脑损伤、缺血性脑损伤、阿尔茨海默病、帕金森病、亨廷顿病、癫痫以及抑郁症等CNS疾病作用的相关研究进展,归纳BDNF在CNS疾病中作用的分子机制,为BDNF用于探索治疗相关CNS疾病的研究提供思路。未来可在实验动物身上探究BDNF的药物动力学机制,通过开发不同的BDNF给药方式及其衍生药物,有望达到治疗CNS相关疾病的效果。

Abstract: This article introduced the important role of brain-derived neurotrophic factor (BDNF) in central nervous system (CNS) function and related diseases, including promoting neuronal growth and survival, enhancing synaptic plasticity, participating in the regulation of neurotransmitters, promoting myelination, and neuroprotective effects, which suggested that BDNF plays an important role in the improvement and treatment of CNS diseases. By summarizing the animal experiments and preclinical studies related to BDNF, the research progress on the effects of BDNF on CNS diseases, such as spinal cord injury, traumatic brain injury, ischemic brain injury, Alzheimer’s disease, Parkinson’s disease, Huntington’s disease, epilepsy and depression, and the molecular mechanism of BDNF in CNS diseases were summarized, which provided ideas for exploring the effects of BDNF in the treatment of related CNS diseases. In the future, the pharmacokinetics mechanism of BDNF can be explored in experimental animals, and different delivery methods of BDNF and its derivatives can be developed to treat CNS related diseases.
