火炬松HMGR基因的系统发育、分子进化和表达分析

Phylogenetic, molecular evolution and expression analyses of HMGR gene in loblolly pine

摘要: 3-羟基-3-甲基戊二酰辅酶A还原酶(HMGR)是植物萜类化合物甲羟戊酸(MVA)生物合成途径的关键酶。为了解其编码基因HMGR的系统发育及其在火炬松进化枝中的分子进化和表达特性。选取19个代表性物种的HMGR基因序列用于系统发育和分子进化分析,并以火炬松5个不同的组织部位为材料分析HMGR基因的表达特性。结果发现:HMGR基因在进化过程中相对保守,序列间的平均遗传距离为0.323,受到的平均选择压大小为0.1212,表明HMGR基因在进化过程主要受纯化选择作用;在火炬松进化枝中受到的选择压大小为0.4293,并有14个位点检测到正向选择作用,且后验概率值大于0.95,表明HMGR基因不同位点受到的选择压差异显著;HMGR基因在火炬松嫩叶中具有最高的表达量,表明HMGR基因的表达具有一定的组织特异性。

Abstract: The 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMGR) is a key enzyme in the MVA biosynthesis pathway of terpenoids. To understand the phylogenetic of its coding gene HMGR and its molecular evolution and expression characteristics in the clade of loblolly pine, in this study, 19 representative species of HMGR sequences were selected for phylogenetic and molecular evolution analysis, and 5 different tissue parts of loblolly pine were used as materials to analyze the expression characteristics of HMGR. The results showed that the HMGR was relatively conserved in the evolution process. The average genetic distance between sequences was 0.323, and the average selection pressure was 0.1212, which indicated that HMGR was mainly subjected to the purifying selection during the evolution process. The selective pressure was 0.4293 in the clade of loblolly pine. Fourteen sites were detected to be positive selective, and the value of posterior probability was greater than 0.95, which indicated that the different selective pressure on each site of HMGR was significant. HMGR had the highest expression level in young leaves of loblolly pine, showing the tissue specific expression of HMGR.
