Rapid extraction and purification of nucleic acid based on magnetic beads

摘要: 采用纳米磁珠法对细菌基因组DNA、总RNA和质粒DNA进行提取纯化,并与试剂盒法和Trizol法提取效果进行比较,以期获得核酸最佳提取方法。结果发现,与试剂盒法和Trizol法相比,磁珠法在细菌基因组DNA和RNA提取、PCR产物检测及纯化中效率更高;而在PCR产物回收和质粒DNA的提取中,效率低于试剂盒法,质粒酶切效果与试剂盒法无差异。结果表明,纳米磁珠法相比试剂盒法和Trizol法在核酸提取纯化效果上更佳。

Abstract: The purpose of this research is to compare the extraction and purification of nucleic acid, including bacterial genomic DNA, plasmid DNA and bacterial total RNA by using the method of magnetic beads and kit and Trizol methods. The results showed that compared with the kit method and Trizol method, the magnetic bead method was more efficient in the extraction of bacterial genomic DNA and RNA, PCR product detection and purification. However, the efficiency of PCR product recovery and plasmid DNA extraction was lower than that of the kit method, and plasmid enzyme cutting effect was not different from that of the kit method. The results showed that the nanometer magnetic bead method is better than the kit and Trizol methods in nucleic acid extraction and purification.
