卡介苗培养滤出液中O-甘露糖基化蛋白的确定和解析

Identification and analysis of O-mannosylated proteins from Bacillus Calmette-Guérin culture filter

摘要: 卡介苗作为预防结核病的唯一疫苗,不能提供全面的保护力,因此全面认识卡介苗的抗原非常关键。甘露糖复合物抗原是存在于分枝杆菌细胞壁上的一大类重要抗原,在细菌毒力以及宿主细胞相互作用等方面发挥作用,其中O-甘露糖基化蛋白在结核分枝杆菌和耻垢分枝杆菌中陆续有报道,但在卡介苗中研究很少。利用Con-A亲和层析技术提纯了卡介苗培养滤出液中的O-甘露糖基化蛋白,并经过ConA-免疫印迹再次确证,最后利用质谱技术进行了解析,共确定出7个O-甘露糖基化蛋白,并对功能和糖基化位点进行了初步预测。O-甘露糖基化蛋白的确定和解析将有助于对卡介苗免疫机制的认识。

Abstract: As the only licensed vaccine to protect against tuberculosis, Bacillus Calmette-Guérin(BCG)can not provide complete protection,therefore, it is essential to fully understand the antigens of BCG. The mannose containing glycoconjugates, which are located on cell wall, play a role in mycobacterial virulence and interaction with host cell. O-mannosylated proteins were reported in Mycobacterium tuberculosis and Mycobacterium smegmatis, but seldom in BCG. In this article, the O-mannosylated proteins from Bacillus Calmette-Guérin culture filter were identified and analyzed by ConA-agarose affinity chromatography combined with MALDI mass spectrometer. There were 7 kinds of O-mannosylated proteins identified and the mannosylated positions of these 7 proteins were further predicted. The identification and analysis of O-mannosylated proteins will shed light on immune mechanism of BCG.
