βGlcY染色技术在枣果实阿拉伯半乳糖蛋白组化定位中的应用

Application of βGlcY dyeing technology in histochemistry localization of arabinogalactan proteins in Ziziphus jujuba Mill cv. Lingwuchangzao

摘要: 以不同发育时期的“灵武长枣”果实为试验材料,应用苯基偶氮染色剂βGlcY染色技术,研究不同发育时期果实阿拉伯半乳糖蛋白(AGPs)的分布特征,并通过免疫荧光定位法进行印证。结果表明:βGlcY-AGPs形成的棕红色沉淀在各时期果实的外果皮及相邻的内部数层排列紧密的中果皮小细胞的细胞壁和细胞内部均有分布。内部的中果皮大型卵圆形薄壁细胞的细胞壁和细胞内在膨大前期均有βGlcY-AGPs形成的棕红色沉淀,而其他时期均主要分布于薄壁细胞的细胞壁上,大部分细胞内部无分布。各时期果实维管束所有细胞的细胞壁和细胞内部都分布有βGlcY-AGPs形成的棕红色沉淀;随果实发育中果皮薄壁细胞等部位βGlcY-AGPs形成的棕红色沉淀逐渐减少,反应AGPs在逐渐减少。同一时期不同浓度βGlcY形成棕红色沉淀的强弱不同,浓度越高染色效果越好;而相同浓度βGlcY在各时期果实不同组织形成的βGlcY-AGPs棕红色沉淀的强弱存在一定差异。βGlcY染色技术是研究灵武长枣果实AGPs组化定位的有效方法。

Abstract: The different developmental stages fruits of Ziziphus jujuba Mill cv. Lingwuchangzao were used as experimental material to study the distribution of arabinogalactan proteins(AGPs) by βGlcY dyeing technology and confirmed by immunofluorescence localization. The main research results were followed: the brown-red precipitate formed by βGlcY-AGPs distributed in the cell walls and cell interiors of the epicarp and the alongside internal layers of closely arranged mesocarp small cells in different periods fruit. The brown-red precipitate formed by βGlcY-AGPs distributed in the cell walls and the inner part of the large ovoid parenchyma cells in the inner mesocarp during the early bulking period, while they were mainly distributed in the parenchyma cell walls during the other periods, with no distribution in the inner part of most cells. The brown-red precipitate formed by βGlcY-AGPs distributed in the cell walls and cell interiors of all cells in the vascular bundles in different periods fruit, the distribution of brown-red precipitate formed by βGlcY-AGPs gradually decreased in the mesocarp parenchyma cells with fruit development, this confirmed that AGPs gradually decreased. The intensity of brown-red precipitate formed by different concentration βGlcY was different at the same periods, the higher the concentration, the better the dyeing effect. There were some differences in brown-red precipitate intensity formed by the same concentration βGlcY in different tissues of fruit at different periods. βGlcY dyeing technology was effective method for AGPs histochemical localization of Ziziphus jujuba Mill cv. Lingwuchangzao fruit.
