Effects of different gut microbes on mating behavior of Drosophila melanogaster

摘要: 通过构建黑腹果蝇的无菌和悉生体系,比较研究不同肠道微生物对无菌果蝇和正常果蝇交配行为的影响。对黑腹果蝇受精卵进行体外消毒处理并在无菌条件下培养获得无菌果蝇。在果蝇培养基中分别接种植物乳杆菌(Lactobacillus plantarum)、醋酸杆菌(Acetobacterium malorum)和酿酒酵母(Saccharomyces cerevisiae),然后对无菌果蝇或正常果蝇分别培养一定时间,获得不同微生物的悉生果蝇。解剖并提取果蝇肠道总RNA,反转录后利用实时荧光定量PCR检测不同微生物在接种前后无菌果蝇和正常果蝇肠道中的丰度,同时对不同处理的果蝇进行单对交配实验,比较不同处理果蝇的交配成功率和交配时间。结果表明:成功构建了无菌果蝇体系,通过与单一微生物共培养,使特定微生物在无菌果蝇和正常果蝇肠道中成功富集,而且植物乳杆菌L. plantarum可通过延长交配潜伏期影响果蝇的交配行为。

Abstract: The germ free and gnotobioticDrosophilamelanogastersystem was constructed, and the effects of different gut microbes on the mating behavior of germ free and conventionalD. melanogasterwere studied. The fertilized eggs ofD. melanogasterwere sterilized in vitro and cultured in sterile condition to obtain germ freeDrosophila.Lactobacillus plantarum,Acetobacterium malorumandSaccharomyces cerevisiaewere inoculated in fly culture medium, respectively, then the germ free and conventional flieswas kept respectively in the culture medium for a certain period of time to obtain different gnotobioticDrosophila.All flies were dissected and the total RNA of the gut was extracted and reverse transcribed, and the abundance of different microbes in the gut of germ free and conventional flies was detected by quantitative real-time PCR (qRT-PCR). Meanwhile, single pair mating assays were carried out to compare the copulation success rate and copulation latency of different treated flies. The results showed that the germ free and gnotobioticDrosophilasystem was successfully constructed, and the specific microbe was successfully enriched in the gut of the germ free and conventional flies after co-cultivation with the single microbe, andL. plantarumcould affect the mating behavior ofDrosophilaby prolonging the copulation latency.
